Anionic Lipids Modulate the Activity of the Aquaglyceroporin GlpF

2015

AbstractThe structure and composition of a biological membrane can severely influence the activity of membrane-embedded proteins. Here, we show that the E. coli aquaglyceroporin GlpF has only little activity in lipid bilayers formed from native E. coli lipids. Thus, at first glance, GlpF appears to not be optimized for its natural membrane environment. In fact, we found that GlpF activity was severely affected by negatively charged lipids regardless of the exact chemical nature of the lipid headgroup, whereas GlpF was not sensitive to changes in the lateral membrane pressure. These observations illustrate a potential mechanism by which the activity of an α-helical membrane protein is modula…

Characterization of the interaction of the antifungal and cytotoxic cyclic glycolipopeptide hassallidin with sterol-containing lipid membranes.

2019

Hassallidins are cyclic glycolipopeptides produced by cyanobacteria and other prokaryotes. The hassallidin structure consists of a peptide ring of eight amino acids where a fatty acid chain, additional amino acids, and sugar moieties are attached. Hassallidins show antifungal activity against several opportunistic human pathogenic fungi, but does not harbor antibacterial effects. However, they have not been studied on mammalian cells, and the mechanism of action is unknown. We purified hassallidin D from cultured cyanobacterium Anabaena sp. UHCC 0258 and characterized its effect on mammalian and fungal cells. Ultrastructural analysis showed that hassallidin D disrupts cell membranes, causin…

Deficiency of glutathione peroxidase-1 accelerates the progression of atherosclerosis in apolipoprotein E-deficient mice.

2007

Background— We have recently demonstrated that activity of red blood cell glutathione peroxidase-1 is inversely associated with the risk of cardiovascular events in patients with coronary artery disease. The present study analyzed the effect of glutathione peroxidase-1 deficiency on atherogenesis in the apolipoprotein E-deficient mouse. Methods and Results— Female apolipoprotein E-deficient mice with and without glutathione peroxidase-1 deficiency were placed on a Western-type diet for another 6, 12, or 24 weeks. After 24 weeks on Western-type diet, double-knockout mice (GPx-1 −/− ApoE −/− ) developed significantly more atherosclerosis than control apolipoprotein E-deficient mice. Moreover…

The selection of aptamers specific for membrane molecular targets

2010

AbstractA growing number of RNA aptamers have been selected experimentally using the SELEX combinatorial approach, and these aptamers have several advantages over monoclonal protein antibodies or peptides with respect to their applications in medicine and nanobiotechnology. Relatively few successful selections have been reported for membrane molecular targets, in contrast to the situation with non-membrane molecular targets. This review compares the procedures and techniques used in selections against membrane proteins and membrane lipids. In the case of membrane proteins, the selections were performed against soluble protein fragments, detergent-membrane protein mixed micelles, whole cells…

Lateral organization of G M1 in phase-separated monolayers visualized by scanning force microscopy

2002

Phase separation of glycolipids in lipid mono- and bilayers is of great interest for the understanding of membrane function. The distribution of the ganglioside GM1 in sphingomyelin (SM)/1-palmitoyl-2-oleoyl- sn-glycero-3-phosphocholine (POPC), SM/1,2-dipalmitoyl- sn-glycero-3-phosphocholine (DOPC) and SM/cholesterol/POPC Langmuir-Blodgett (LB) monolayers transferred at 36 mN/m has been studied by scanning force microscopy. Besides lateral organization of the glycolipid in LB monolayers as deduced from topography, material properties have been investigated by phase imaging, pulsed force mode and force modulation microscopy. It was shown that GM1 preferentially clusters in an ordered lipid m…

Effect of temperature on the growth of alfa-PbO2 nanostructures

2010

Abstract Ordered arrays of α-PbO 2 nanostructures were grown by galvanostatic anodic deposition into the channels of alumina templates. Electrodepositions were performed in an aqueous solution containing lead acetate and sodium acetate at pH 5.4. Bath temperature and electrodeposition time were varied to check their effect on the growth of nanostructures. It has been found that filling of alumina pores is independent of the time and electrodeposition temperature, whilst height and growth kinetics of nanostructures vary with both parameters. Temperature greatly influences morphology: wires grown at room temperature consisted of clusters of particles, leading to poorly compact structures, whi…

Current‐voltage curves of bipolar membranes

1992

Bipolar membranes consist of a layered ion‐exchange structure composed of a cation selective membrane joined to an anion selective membrane. They are analogous to semiconductor p‐n devices as both of them present current‐voltage curves exhibiting similar rectification properties. In this article, we present some current‐voltage curves obtained for different bipolar membranes at several temperatures. The results can be interpreted in terms of a simple model for ion transport and field‐enhanced water dissociation previously developed. The mechanism responsible for water splitting is assumed to be a catalytic proton transfer reaction between the charged groups and the water at the membrane int…
