Postmortem distribution of dihydrocodeine and metabolites in a fatal case of dihydrocodeine intoxication.

A report of a fatal dihydrocodeine ingestion under substitution therapy is given. Quantitation of dihydrocodeine, dihydromorphine, N-nordihydrocodeine, dihydrocodeine-6-, dihydromorphine-6- and dihydromorphine-3-glucuronide was performed simultaneously after solid-phase extraction prior to HPLC analysis, and the analytes were detected using their native fluorescence. Postmortem concentrations of blood samples from different sampling sites as well as from liver, kidney and cerebrum are reported. The use of fish bile metabolite analyses as exposure biomarkers to pulp and paper mill effluents

The exposure of caged one-year-old whitefish (Coregonus lavaretus) to wood extractives and chlorophenolics from pulp and paper mill effluents in Lake Saimaa (SE Finland) was investigated. Whitefish were exposed at 17 locations affected by effluents from pulp and paper mills and at 5 reference areas. Resin and fatty acids and chlorophenolics in bile were analysed by GC and GC-MS. Cholesterol concentration in bile was also measured. : The gut microbial metabolite, trimethylamine N-oxide (TMAO), was previously reported to induce platelet hypersensitivity, which leads to thrombotic risk. However, the molecular mechanism underlying the effects of TMAO on endothelial cells (EC), which is the primary vessel wall contact with the lumen, remains unclear. Here, we investigated the impact of TMAO on procoagulant activity (PCA) in EC and mice, for a possible link between microbiota and coagulation. Abstract Phytoremediation technologies such as constructed wetlands have shown higher efficiencies in removal of pharmaceuticals from wastewaters than conventional wastewater treatment processes, and plants seem to have an important role in the removal of some of those compounds. In this context, a study was conducted to assess tolerance, uptake, and metabolism of the epilepsy drug, carbamazepine, by the macrophyte Typha spp. This evaluation was conducted in hydroponic solutions with 0.5–2.0 mg/L of this pharmaceutical for a maximum period of 21 days. The phosphorylation of the 2 major phenobarbital-inducible cytochrome P450 isoenzymes IIB1 and IIB2 was increased in intact hepatocytes by the action of the membrane-permeating cAMP derivative N6,O2'-dibutyryl-cAMP. Under these conditions cyclophosphamide and ifosfamide (which are known to be activated by cytochrome P450 IIB1) were investigated for mutagenicity in Salmonella typhimurium TA1535 and TA100 and for cytotoxicity in TA1535. Cyclophosphamide and ifosfamide were transformed to mutagenic and cytotoxic metabolites by the hepatocytes. A method has been developed to separate hydroxylated metabolites of the carcinogenic polycyclic aromatic hydrocarbon benzo[a]pyrene, i. e. trans-4,5-, 7,8-, 9,10-dihydrodiol and 1-, 3-, 7-, and 9-phenol, by HPLC with amperometric detection employing an isocratic methanol/water eluent (70:30, v/v) containing 0.5 g/L sulfuric acid and 1 g/L lithium perchlorate. Based on the natural abundance of 13C, in vivo13C-NMR was used for the first time to monitor the metabolism of sucrose and hydroquinone (1) in cell suspensions of the plant Rauwolfia serpentina (L.) BENTH. ex KURZ. Cells converted sucrose extracellularly into α-D- and β -D-glucose as well as into β -D-fructofuranose and β -D-fructopyranose, respectively. The sugar mixture was completely taken up by the cells after 4 days. Hydroquinone fed at that time resulted in optimum conversion into its β -D-glucoside arbutin (2) within 10 h.
