Roles of EGFR and KRAS and their downstream signaling pathways in pancreatic cancer and pancreatic cancer stem cells

2015

Pancreatic cancer is currently the fourth most common cancer, is increasing in incidence and soon will be the second leading cause of cancer death in the USA. This is a deadly malignancy with an incidence that approximates the mortality with 44,000 new cases and 36,000 deaths each year. Surgery, although only modestly successful, is the only curative option. However, due the locally aggressive nature and early metastasis, surgery can be performed on less than 20% of patients. Cytotoxic chemotherapy is palliative, has significant toxicity and improves survival very little. Thus new treatment paradigms are needed desperately. Due to the extremely high frequency of KRAS gene mutations (>90%) d…

GENETIC AND FUNCTIONAL DISSECTION OF THE MIR-17-92 CLUSTER OF MIRNAS

2011

Background: The miR-17∼92 cluster was among the first cluster of miRNAs to be linked to cancer. It is directly activated by the members of the MYC family of transcription factors and acts as a bona fide oncogene in several types of cancer, including lymphomas. miR-17∼92 is essential for mammalian development. Knockout mice die in utero or at birth with cardiovascular defects and impaired B-cell maturation. Aims: Although miR-17∼92 is important for tumorigenesis and development both the molecular mechanisms and the prominent miRNAs acting in different contexts are still unknown. The main aim of our study was to identify the role played by different members of the miR-17∼92 cluster in B-cell …

Classification trees for preference data: a distance-based approach

2014

In the framework of preference rankings, when the interest lies in explaining which predictors and which interactions among predictors are able to explain the observed preference structures, the possibility to derive consensus measures using a classi cation tree represents a novelty and an important tool given its easy interpretability. In this work we propose the use of a multivariate decision tree where a weighted Kemeny distance is used both to evaluate the distances between rankings and to de ne an impurity measure to be used in the recursive partitioning. The proposed approach allows also to weight di erently high distances in rankings in the top and in the bottom alternatives.

Enhanced volcanic hot-spot detection using MODIS IR data: results from the MIROVA system

2015

We describe a new volcanic hotspot detection system, named Middle InfraRed Observation of Volcanic Activity (MIROVA), based on the analysis of infrared data acquired by the Moderate Resolution Imaging Spectroradiometer sensor (MODIS). MIROVA uses the middle infrared radiation (MIR), measured by MODIS, in order to detect and measure the heat radiation deriving from volcanic activity. The algorithm combines spectral and spatial principles, allowing the detection of heat sources from 1 megawatt (MW) to more than 10 gigawatt (GW). This provides a unique opportunity to: (i) recognize small-scale variations in thermal output that may precede the onset of effusive activity; (ii) track the advance …

A role for miR-142-3p in colony-stimulating factor 1-induced monocyte differentiation into macrophages

2013

AbstractThe differentiation of human peripheral blood monocytes into macrophages can be reproduced ex vivo by culturing the cells in the presence of colony-stimulating factor 1 (CSF1). Using microarray profiling to explore the role of microRNAs (miRNAs), we identified a dramatic decrease in the expression of the hematopoietic specific miR-142-3p. Up- and down-regulation of this miRNA in primary human monocytes altered CSF1-induced differentiation of monocytes, as demonstrated by changes in the expression of the cell surface markers CD16 and CD163. One of the genes whose expression is repressed by miR-142-3p encodes the transcription factor Early Growth Response 2 (Egr2). In turn, Egr2 assoc…

Ierogamie, euritmie e ierotopie mariane in Sicilia.

What…

How miR-31-5p and miR-33a-5p Regulates SP1/CX43 Expression in Osteoarthritis Disease: Preliminary Insights

2021

Osteoarthritis (OA) is a degenerative bone disease that involved micro and macro-environment of joints. To date, there are no radical curative treatments for OA and novel therapies are mandatory. Recent evidence suggests the role of miRNAs in OA progression. In our previous studies, we demonstrated the role of miR-31-5p and miR-33a families in different bone regeneration signaling. Here, we investigated the role of miR-31-5p and miR-33a-5p in OA progression. A different expression of miR-31-5p and miR-33a-5p into osteoblasts and chondrocytes isolated from joint tissues of OA patients classified in based on different Kellgren and Lawrence (KL) grading was highlighted
