In the context of radiopharmacy and molecular imaging, the concept of theranostics entails a therapy-accompanying diagnosis with the aim of a patient-specific treatment. Using the adequate diagnostic radiopharmaceutical, the disease and the state of the disease are verified for an individual patient. 72/74As-labeling of HPMA based polymers for long-term in vivo PET imaging

Abstract In the context of molecular imaging, various polymers based on the clinically approved N-(2-hydroxypropyl)-methacrylamide (HPMA) have been radio-labeled using longer-living positron emitters 72As t1/2 = 26 h or 74As t1/2 = 17.8 d. This approach may lead to non-invasive determination of the long-term in vivo fate of polymers by PET (positron emission tomography). Presumably, the radio label itself will not strongly influence the polymer structure due to the fact that the used nuclide binds to already existing thiol moieties within the polymer structure. Thus, the use of additional charges or bulky groups can be avoided.

Generator-derived radionuclides for PET/CT imaging are promising for optimizing targeted radiotherapy by an individual patient-based approach, applying pre-therapeutic evaluation, dosimetric calculations, and for measuring treatment response after radionuclide therapy. To realize this vision, sophisticated nuclear chemistry and radiopharmaceutical chemistry should coincide with quantitative molecular imaging and dosimetric calculations.

Efficient gene therapy based targeting system for the treatment of inoperable tumors

Background A considerable percentage of tumors are not amenable to surgery. We have designed a simple and powerful targeting system that offers an alternative option for the multi-component pre-targeting strategies used clinically. This targeting system can be used for any type of solid tumors independent of the tumor type, thereby omitting the need to engineer unique antibodies for each specific application or tumour type. Development of MR active contrast agents via Parahydrogen Induced Polarization

Parahydrogen Induced Polarization provides dramatic MR signal enhancement that can be exploited for molecular imaging. This method allows amongst others for Magnetic Resonance Imaging of 13C and 15N, which is usually constrained by the low MR sensitivity of these nuclei. By combining hydrogenation of barbiturates with parahydrogen under special experimental conditions (PASADENA under pressure) with a polarization transfer sequence we demonstrate the transfer of the initial 1H polarization to 13C.
