Cardiotoxicity of mitoxantrone treatment in a german cohort of 639 multiple sclerosis patients

2014

Background and PurposezzThe aim of this study was to elucidate the role of therapy-related cardiotoxicity in multiple sclerosis (MS) patients treated with mitoxantrone and to identify potential predictors for individual risk assessment. MethodszzWithin a multicenter retrospective cohort design, cardiac side effects attributed to mitoxantrone were analyzed in 639 MS patients at 2 MS centers in Germany. Demographic, disease, treatment, and follow-up data were collected from hospital records. Patients regularly received cardiac monitoring during the treatment phase. ResultszzNone of the patients developed symptomatic congestive heart failure. However, the frequency of patients experiencing car…

Cardio-oncology in multiple myeloma: is it time for a specific focus?

2017

It has been known for some time that some oncologic drugs can cause heart damage. The term ‘cardiotoxicity’ just refers to the presence of a cardiac event during therapy or at least related to ther...

Indefinite integrals involving complete elliptic integrals of the third kind

2017

ABSTRACTA method developed recently for obtaining indefinite integrals of functions obeying inhomogeneous second-order linear differential equations has been applied to obtain integrals with respect to the modulus of the complete elliptic integral of the third kind. A formula is derived which gives an integral involving the complete integral of the third kind for every known integral for the complete elliptic integral of the second kind. The formula requires only differentiation and can therefore be applied for any such integral, and it is applied here to almost all such integrals given in the literature. Some additional integrals are derived using the recurrence relations for the complete …

Carotid restenosis is associated with plasma ADMA concentrations in carotid endarterectomy patients

2011

Abstract Background: The aim of this work has been to study the association between plasma asymmetric dimethylarginine (ADMA) concentrations and carotid stenosis in a group of 64 patients undergoing carotid endarterectomy (CEA). Methods: Arginine, ADMA and symmetric dimethylarginine (SDMA) were measured using capillary electrophoresis with UV detection. An evaluation of plasma concentrations of total cysteine (tCys) and total homocysteine (tHcy) was also performed. Results: Pearson’s analysis show a positive correlation between ADMA and carotid stenosis (r=0.37, p=0.003), which is also confirmed after stepwise multiple linear regression analysis. ADMA plasma concentrations were significantl…

Generation of soliton molecules, power regulation, regeneration and sculpting of pulse profiles within a multifunction fiber laser

2018

The work of this thesis strives to provide a solution to the persistent problem of poverty of mode-locked fiber lasers, in terms of functionality and flexibility. The thesis proposes a fiber-laser cavity having the specificity of being multifunctional. The cavity is equipped with tunable components, which provide the flexibility to realize functions ranging from the generation of pulses with complex profiles (solitons, bi-solitons, tri-solitons, etc.) up to the carving of pulse profiles, passing through the regulation of peak powers, and the regeneration of severely degraded intensity profiles. The laser cavity that we propose has the specificity of being controlled by a key component, whic…

Direct determination of intracellular daunorubicin in intact confluent monolayers of AT1 prostate carcinoma cells using a multiwell–multilabel counter

2008

The cytostatic drug daunorubicin exerts its toxic action by intercalating into the DNA. The efficacy of daunorubicin depends on the intracellular amount in the tumor cell. Here we have evaluated the use of a multiwell-multilabel reader for the direct determination of the fluorescent cytostatic drug daunorubicin in a prostate carcinoma cell line (AT1 R-3327 Dunning prostate carcinoma cells) grown on 24-well plates. We present evidence that this simple fluorescent parameter is a good measure for the toxicologically relevant amount of the drug intercalated into the DNA and, therefore, is a good predictor for the drug's cytotoxicity. The amount of cationic cytostatics in a tumor cell is primari…

New artesunic acid homodimers: Potent reversal agents of multidrug resistance in leukemia cells

2012

Abstract To evade the problem of multidrug resistance, hybridization of natural products in dimers is considered as an effective method. After the successful synthesis of three artesunic acid homodimers connected by different types of chemical linkers, we analyzed their activity against human CCRF-CEM and multidrug-resistant p -glycoprotein-overexpressing CEM/ADR 5000 leukemia cells and observed, that multidrug resistant cells were not cross-resistant to the new compounds. Collateral sensitivity was observed for artesunic acid homodimer 2. The obtained results deliver valuable information about the linker’s structure which is required for homodimers to be highly cytotoxic.

Synthesis and biological evaluation of a D-ring-contracted analogue of lamellarin D

2017

A D-ring contracted analogue of the strongly cytotoxic marine pyrrole alkaloid lamellarin D was synthesized and investigated for its antiproliferative action towards a wild type and a multidrug resistant (MDR) cancer cell line. The compound was found to inhibit tumor cell growth at submicromolar concentrations and showed a lower relative resistance in the MDR cell line than the antitumor drug camptothecin to which lamellarin D shows cross resistance and with which lamellarin D shares the same binding site.

Targeting multiple myeloma with natural polyphenols

2019

Multiple myeloma (MM) is still an incurable hematologic malignancy. Although new therapeutic strategies have been developed to target different pathways in malignant cells, such as proliferation, differentiation, and apoptosis, better survival rates have also been achieved by the introduction of autologous stem cell transplantation (ASCT). Hematopoietic stem cell transplantation and novel targeted agents, such as proteasome inhibitors, monoclonal antibodies, immunomodulatory drugs, check-point inhibitors and epigenetic modulators, have significantly achieved long remission time and increased survival rates. However, most patients relapse, develop resistance, and eventually die because of re…

Antitumor effects of curcumin and structurally β-diketone modified analogs on multidrug resistant cancer cells

2007

Abstract Using concepts of bioisostery a series of curcumin analogs were synthesized: the diketonic system of the compound was elaborated into enaminones, oximes, and the isoxazole heterocycle. The cell growth inhibitory and apoptosis inducing effects of the new analogs were evaluated by in vitro assays in the hepatocellular carcinoma HA22T/VGH cells, as well as in the MCF-7 breast cancer cell line and in its multidrug resistant (MDR) variant MCF-7R. Increased antitumor activity on all cell lines was found with the isoxazole analog and especially with the benzyl oxime derivative; in the HA22T/VGH cell model, the latter compound inhibited constitutive NF-κB activation.
