Antiabsence effects of carbenoxolone in two genetic animal models of absence epilepsy (WAG/Rij rats and lh/lh mice).

2005

Carbenoxolone (CBX), the succinyl ester of glycyrrhetinic acid, is an inhibitor of gap junctional intercellular communication. We have tested its possible effects upon two genetic animal models of epilepsy (WAG/Rij rats and lethargic (lh/lh) mice). Systemic administration of CBX was unable to significantly affect the occurrence of absence seizures in WAG/Rij rats. In particular, intravenous (5-40 mg/kg) or intraperitoneal (i.p.; 10-80 mg/kg) administration of CBX was unable to significantly modify the number and duration of spike-wave discharges (SWDs) in WAG/Rij rats, whereas the bilateral microinjection (0.05, 0.1, 0.5 and 1 microg/0.5 microl) of CBX into nucleus reticularis thalami (NRT)…

Transcriptional expression of selected genes associated with excretion of carboxylic acids from aci mutants of Saccharomyces cerevisiae

2013

Introduction: Saccharomyces cerevisiae is an excellent model organism for studies of transcriptional regulation of metabolic processes in other eukaryotic cells including human cells. Cellular acid-base balance can be disturbed in pathologic situations such as renal acidosis or cancer. The extracellular pH of malignant solid tumors is acidic in the range of 6.5-6.9. EG07 and EG37 aci mutants of Saccharomyces cerevisiae excessively excrete carboxylic acids to glucose-containing media or distilled water. The excreted acids are Krebs and/or glyoxylate cycle intermediates. The genes restoring the wild-type phenotype have function that does not easily explain theAci phenotype.Material/Methods: I…

Perturbed interactions of mutant proteolipid protein/DM20 with cholesterol and lipid rafts in oligodendroglia: implications for dysmyelination in spa…

2006

Missense mutations in the humanPLP1gene lead to dysmyelinating diseases with a broad range of clinical severity, ranging from severe Pelizaeus–Merzbacher disease (PMD) to milder spastic paraplegia type 2 (SPG-2). The molecular pathology has been generally attributed to endoplasmic reticulum (ER) retention of misfolded proteolipid protein (PLP) (and its splice isoform DM20) and induction of the unfolded protein response. As opposed to previous studies of heterologous expression systems, we have analyzed PLP/DM20 trafficking in oligodendroglial cells, thereby revealing differences between PMD and SPG-2-associated PLP/DM20 isoforms. PLPA242Vand DM20A242V(jimpy-msdin mice), associated with seve…

MR3257881 Reviewed Hadwin, Don Approximate double commutants in von Neumann algebras and C∗-algebras. Oper. Matrices 8 (2014), no. 3, 623–633. (Revie…

2015

In this paper, the author proves an asymptotic version of the double commutant theorem, in a particular set-up of commutative C∗ -algebras. More precisely, he considers the relative approximate double commutant of a C ∗-algebra with unit, and, using a theorem of characterization for a commutative C∗-subalgebra with unit (inspired by a well-known result due to Kadison for a von Neumann sub-algebra of type I), and from a theorem based on a Machado result, he proves that if A is a commutative C∗-subalgebra of a C∗-algebra B centrally prime with unit, then A is equal to its relative approximate double commutant. In the case where B is a von Neumann algebra, a distance formula is found.

Study of the role of “gatekeeper” mutations V654A and T670I of c-kit kinase in the interaction with inhibitors by means mixed molecular dynamics/dock…

2011

The over-expression of c-kit proto-oncogene has been reported in hematopoietic cells, small cell lung cancer, and gastrointestinal stromal tumors. The clinical importance of c-kit expression in tumors focused the research towards inhibitors of this tyrosine kinase. Imatinib (Gleevec®) was the first compound used in therapy, but mutations on c-kit led to reduced effectiveness or ineffectiveness of this treatment. Other compounds are likely to be effective against mutants, such as Sunitinib (Sutent®), but the need for new and most effective inhibitors against mutants is still critical. We report mixed Molecular Dynamics/Docking study with the aim to unveil the molecular mechanism involved in …

Endosperm cintribution to Medicago truncatula seed development : characterization of a DOF transcription factor expressed in chalazal endosperm

2014

In the current context, which necessitates a reduction in inputs in crop systems and boosting of production of plant proteins to reduce France’s dependency on feed imports,, growing legumes represents an alternative. Grain legumes are major sources of proteins for animal and human nutrition. In the UMR1347 Agroécologie, the objectives of the study group "déterminismes Génétiques et Environnementaux de l’Adaptation des Plantes à des Systèmes de culture Innovants" (GEAPSI) are to promote legume cultivation and adaptation to environmental stresses, via multidisciplinary approaches (genetics, ecophysiology, molecular physiology). This thesis project was carried out in the "Étude des Mécanismes …

Identification and transcriptional profile of Lactobacillus paracasei genes involved in the response to desiccation and rehydration

2020

International audience; Lactobacillus paracasei is able to persist in a variety of natural and technological environments despite physico-chemical perturbations, in particular alternations between desiccation and rehydration. However, the way in which it adapts to hydric fluctuations and the genetic determinants involved are not clearly understood. To identify the genes involved in adaptation to desiccation, an annotated library of L. paracasei random transposon mutants was screened for viability after desiccation (25% relative humidity, 25 °C). We found 16 genes that have not been described as being involved in this response. Most of them are linked to either the transport of molecules or …

Resistance to virus infection mediated by artificial microRNAs: estimating the likelilhood of escape mutants

2013

Short term adaptive response of symbiotic N2 fixation in pea to root pruning of half the root system, linked to the availability of carbon assimilates

2014

Symbiotic N fixation of legumes is very sensitive to environmental stresses, like pea pests damaging nodulated roots. However, the impact on their N uptake capacity and plant growth has not been studied so far.We analyzed the adaptive response symbiotic N2 fixation and plant growth of pea wild type Frisson and hypernodulating mutants P64, P118 and P121 mutated respectively on genes SYM28, SYM29 and NOD3 to root pruning of half the root system at the end of the vegetative stage. The adaptive responses of pea: cv. Frisson and 3 of its hypernodulating mutants were compared under varying carbon supplies from photosynthesis. At 380 ppm, mutant P118 showed the lowest decrease of the specific activ…
