IAPs and Resistance to Death Receptors in Cancer

Since their identification in mammal cells, IAPs emerged have as potent regulators of death receptor signalling pathways, determining the cell fate in response to receptor stimulation. Among IAPs, cIAP1 and cIAP2 are active components of receptor-associated signalling complexes able to promote the activation of ubiquitin-dependent survival signalling pathways. For its part, XIAP is an important regulator of caspase activity, determining the apoptotic signalling pathway engaged after death receptor stimulation. The use of IAP antagonists is a promising strategy in order to overcome the resistance of tumor cells to death receptor stimulation.
H2O2 INDUCES NECROPTOSIS IN MESOANGIOBLAST STEM CELLS

Stem cells are used in regenerative medicine, but their therapeutic efficacy is compromised by their huge death during the first days post-transplantation. Indeed, the microenvironment within damaged tissues is hostile for stem cell survival mainly due to oxidative stress. H2O2 may play a relevant role in inducing death of the injected cells. The aim of our study was to determine the mechanism of mesoangioblast (A6) cell death after an H2O2 treatment. Targeting NUPR1 with the Small Compound ZZW-115 Is an Efficient Strategy to Treat Hepatocellular Carcinoma

International audience; HCC is a highly lethal malignancy with Sorafenib as the only molecularly targeted drug. The multifunctional stress-associated protein, NUPR1, plays an essential role in controlling cell growth, migration, invasion and Sorafenib resistance in HCC. We report here that NUPR1 expression is absent in healthy liver and it is progressively upregulated in HCC premalignant lesions such as hepatitis and cirrhosis with a maximum expression in HCC samples, highlighting that NUPR1 is a potential drug target for HCC.
