Cyclooxygenase-1/2 (COX-1/COX-2) and 5-lipoxygenase (5-LOX) inhibitors of the 6,7-diaryl-2,3-1H-dihydropyrrolizine type

A series of 6,7-diaryl-2,3-1H-dihydropyrrolizines was prepared as COX-1/COX-2 and 5-LOX inhibitors. The inhibition of COX-1 was evaluated using intact bovine platelets as the enzyme source, whereas LPS-stimulated human monocytes served as the enzyme source for inducible COX-2. The determination of arachidonic metabolites was performed by HPLC for COX-1 and RIA for COX-2. The balance between COX-1/COX-2 and 5-LOX inhibition can be shifted by modifying the substitution pattern of the phenyl moiety at the 6- and 7-position of the pyrrolizine nucleus. Structure-activity relationships are discussed.

Effects of motor cortex removal on circling behaviour.

AbstractChemical stimulation of the dopaminergic nigro-striatal system induces circling behaviour also in animals in which an extensive telencephalic lesion has been performed. In order to avoid the effects linked to chemical stimulation, we have studied the circling behaviour means of electrical stimulation of the caudate nucleus in chronic cats with cortical lesions. The role of the motor cortex on the circling behaviour is emphasized.

Abstract Cannabis sativa (hemp) is a flowering annual plant whose phytochemical by-products, hashish and marihuana, are the most widely produced and most frequently used illicit drugs in Europe. Δ 9 -Tetrahydrocannabinol is the primary psychoactive constituent, responsible, in a dose-related manner, for euphoria, cognitive effects, and psychotic symptoms, as well as the addictive potential of smoked cannabis due to its interference with the mesolimbic dopaminergic system. Tumors of endothelial origin develop rarely. Until now, only two angiosarcoma (AS)-derived endothelial cell lines have been be isolated, ISO-HAS and AS-M. Both AS-derived endothelial cell lines presented the typical endothelial characteristics, such as the expression of CD31 and von Willebrand factor, but differed from normal endothelial cells in a nuclear expression of p53, in a delayed angiogenic reaction, and a reduced expression of caveolin. In addition, differences in the expression of cytokines and cell adhesion molecules responsive to proinflammatory stimuli were observed. The effect of triiodothyronine (T3) on the rate of synthesis of nuclear proteins was studied during terminal differentiation of rat cortical neurons cultured in a serum-free medium. To this aim total and acid soluble nuclear proteins were analyzed by different electrophoretic techniques. Our results show that: 1) during maturation in vitro, neuronal nuclei undergo a dramatic change in the rate at which different classes of histones and high mobility group (HMG) proteins are synthesized; the synthetic activity, measured as incorporation of radioactive precursors into nuclear proteins, slows indeed down with age: especially evident is the decrease in core histones synthesis; at day 15, on the…
