Valorization of natural drug products : from extraction to encapsulation

This thesis was carried out at the Laboratory of Pharmacognosy and the Laboratory of Pharmaceutical technology, at the UFR Sciences de Santé, circonscription Pharmacy, in the University of Burgundy Franche-Comté, to find new natural molecules to encapsulate. First of all, we focused on the natural saponins from plants of the Vietnam biodiversity and the french horticulture, belonging to the three genera Dracaena, Cordyline (Asparagaceae) and Weigela (Caprifoliaceae). The work led to the successful isolation and elucidation of 42 natural saponins using various chromatographic techniques. The structures were determined by ESI mass spectrometry and NMR spectroscopy. X-ALD and P-NALD are two rare, peroxisomal metabolic and neurodegenerative diseases. ABCD1 and ACOX1 are known to be responsible for X-ALD and P-NALD, respectively. The actively demyelinating lesions in CNS, exhibited signs of oxidative stress and cell death. The accumulation of VLCFA in plasma and tissue is the biochemical common hallmark to both diseases. First, we characterized a murine oligodendrocytes cell line 158N to use it as a model. This thesis aims to develop an edible packaging made of a mixture of chitosan and gelatin (beef or fish), to better understand the influence of the electron beam irradiation and of the incorporation of natural antioxidants on the physico-chemical and functional properties of the films. A study of the effect of irradiation on antioxidants release kinetics in a simple liquid medium was studied for validation. Despite the development of new anti-cancer drugs such as anti-tyrosine kinases or anti-angiogenic therapy, cancer mortality remains high. These new molecules associated with advances in cancer surgery, radiotherapy and chemotherapy have succeeded in improving life expectancy in these patients. It has also allowed a long-term evaluation of the cardiovascular impact of these therapies. Many chemotherapy drugs, such as anthracyclines, lead to impaired cardiac function. The aim of this work was to select rationally a probiotic strain by setting up a screening method as well as study and understand the impact of the redox potential (Eh) and gas bubbling on the survival of Bifidobacterium bifidum in a fermented dairy product. A concept that has emerged in recent years is the use of micronutrients as therapeutic agents known for their anti-inflammatory, antioxidant, cardio-protective or anticarcinogenic proprieties. In this work, we studied the in vivo and in vitro mechanism of action of resveratrol and two natural formulations, one involved in the antioxidant effects (product "X") and the other (product "Y") involved in the correction of lipid disorders in people affected by metabolic syndrome. Diabetes and insulin resistance are associated with the development of cardiovascular and nervous diseases.
