Cytometric analysis for drug-induced steatosis in HepG2 cells

2009

Drugs are capable of inducing hepatic lipid accumulation. When fat accumulates, lipids are primarily stored as triglycerides which results in steatosis and provides substrates for lipid peroxidation. An in vitro multiparametric flow cytometry assay was performed in HepG2 cells by using fluorescent probes to analyze cell viability (propidium iodide, PI), lipid accumulation (BODIPY493/503), mitochondrial membrane potential (tetramethyl rhodamine methyl ester, TMRM) and reactive oxygen species generation (ROS) (2',7'-dihydrochlorofluorescein diacetate, DHCF-DA) as functional markers. All the measurements were restricted to live cells by gating the cells that excluded PI or those that exhibited…

Peroksīdu noteikšana ar eksprestestu metodi

2017

Peroksīdu noteikšana ar eksprestestu metodi. Bruņeniece A., darba vadītājs Dr. ķīm. docents Ģībietis J. Bakalaura darbs, 44 lappuses, 10 tabulas, 26 attēli, 23 literatūras avoti. Latviešu valodā. Darbā apskatītas dažādas ūdeņraža peroksīda un peroksietiķskābes kvantitatīvās noteikšanas metodes. Veikta ITS firmas ekprestesta „Water-Works” teststrēmeļu kalibrēšana. Eksperimentāli izpētīta jonu spēka, pH un organiskā šķīdinātāja ietekme uz teststrēmeļu darbību. Iegūti kalibrēšanas grafiki un izveidotas krāsu standartskalas, izmantojot spektrofotometru – kolorimetru AvaMouse Avantes, kā arī izpētītа teststrēmeļu piеmērotība peroksīdu koncentrācijаs noteikšanаi atšķirīgās vidēs. Indium doping of proton-conducting solid oxides

2008

Solid oxides protonic conductors are prepared by doping the pure matrix compounds with cationic species. Barium cerate and barium zirconate are perovskite-like compounds, characterized by a network of corner-sharing MeO6 octahedra (Me=Ce, Zr). Barium lies in the cavities between octahedra. Insertion of trivalent species in the octahedral site involves the formation of charge- compensating oxygen vacancies, that can be filled by hydroxyls coming from dissociative water absorption. Then, proton delocalization among structural oxygens ensures conductivity. The most effective conductors are obtained by yttrium doping that, on the other hand, enters only in limited amounts in both BaZrO3 and BaC…

Toddlers' Play in Early Childhood Education Settings

2018

More and more toddlers participate in early childhood education outside home, sharing a significant part of their day with teachers and peers, often in play. Despite the knowledge that has accumulated on toddlers’ playing skills and on the psychology underlying the development of play, research on toddlers’ play from the educational and pedagogical viewpoints is limited. Whereas teachers of toddlers report high respect for play and the importance of play in toddlers’ lives, research shows that the position of play is generally weak. This chapter, drawing on existing research, reviews the development and characteristics of toddlers’ play, describes the ways in which they play, and discusses …

Environmentally driven changes in Baltic salmon oxidative status during marine migration.

2020

The fitness and recruitment of fish stocks can be markedly affected by environmental disturbances including global warming, eutrophication and contamination. Understanding the effects of environmental stressors on salmon physiology during marine residence is of a global concern as marine survival has decreased. We present a unique combination of physiological responses - antioxidant defence and oxidative damage biomarkers, stable isotopes and contaminant exposure biomarkers - measured from adult Atlantic salmon (Salmo salar) collected at the Baltic Sea and studied in relation to environmental variables and fitness estimates. The results demonstrate that feeding populations of salmon display…

Scaffolding patterns of dialogic exchange in toddler classrooms

2021

The present study investigates how often episodes of dialogic exchange can be identified in educator–toddler interactions during play and emerging academic activities, what kind of scaffolding strategies educators use during the episodes of dialogic exchange and, ultimately, what kind of scaffolding patterns of dialogic exchange can be identified based on educators' use of scaffolding strategies. Educator–child interactions were video-recorded in seven European countries, in two toddler classrooms in each country. The video recordings of emerging academic and play activities were transcribed and analysed, combining the principles of theory and a data-driven content analysis. First, 69 episo…

Impairment of plasma nitric oxide availability in senescent healthy individuals: Apparent involvement of extracellular superoxide dismutase activity

2006

To verify the potential involvement of the age-dependent modifications of EC-SOD activity in the impairment of plasma NO availability with advancing age, 40 healthy men divided into 4 age groups for the purpose of comparison (young: 27.4 +/- 1.5 years; middle: 50.8 +/- 2.2, years; old: 70.0 +/- 1.8 years; very old: 86.1 +/- 1.1 years) were enrolled in this study. Plasma samples were used for measurements of the stable end-product nitrite/nitrate (NOx), as an expression of NO availability, EC-SOD activity, thiobarbituric acid reactive substances (TBARS) as a marker of lipid peroxidation, low density lipoprotein (LDL) copper-mediated oxidation in vitro and total antioxidant capacity (TEAC). Point-Of-Care Plastic Biosensor Devices using a PEDOT-modified Electrochemical Glucose Sensing Platform

2015

There is an emerging global epidemic of diabetes that can be related to rapid increases in overweight, including obesity and physical inactivity. According to World Healthcare Organization, diabetes is a leading cause of blindness, amputation and kidney failure, while total deaths from diabetes are projected to rise by more than 50% in the next 10 years. To prevent life threatening and debilitating complications associated with diabetes, researchers are continuously involved in developing glucose biosensors. Hence, it is essential to develop a biosensor that can determine glucose levels having fast-response, high sensitivity and selectivity. Depending upon the transduction technique, a numb…
