Changes in the proton potential and the cellular energetics of Escherichia coli during growth by aerobic and anaerobic respiration or by fermentation.

The energetic parameters of Escherichia coli were analyzed for the aerobic/anaerobic transition. The electrochemical proton potential (delta p) across the cytoplasmic membrane was determined in the steady state of respiration with O2, nitrate, fumarate, dimethylsulfoxide (Me2SO), and for fermentation. With O2, a proton potential of -160 mV was obtained. For anaerobic respiration with nitrate, fumarate or Me2SO, delta p decreased only slightly by about 20 mV in contrast to earlier assumptions, whereas delta p dropped by approximately 40 mV during fermentation. The reaction in water of cobalt(II), nickel(II) or manganese(II) chloride with 1,10-phenanthroline (phen) and sodium pyrophosphate (Na 4 P 2 O 7 ) at low pH (∼2) afforded three isostructural pyrophosphate complexes of an unprecedented one-dimensional typology, namely, {[M(phen)(H 2 O)(H 2 P 2 O 7 )]·H 2 O} n with M = Co( 1 ), Ni( 2 ) and Mn( 3 ). The di-hydrogen-pyrophosphate anion featured in these complexes adopts a rare bidentate/monodentate bridging mode leading to chain propagation. This unusual bridging pathway produces a metal–metal intra-chain separation of about 6.6–6.7 A for 1 – 3 , values much larger than the metal–metal distance across the classic bis-bidentate PPi in the parent…

The new ternary compounds UP2S6, UP2S7, U(P2S6)2, and U3(PS4)4 were prepared from uranium metal, phosphorus pentasulfide, and sulfur at 700 °C. The crystal structures were determined by single-crystal X-ray diffraction methods. UP2S6 (I) crystallizes in the ZrP2S6 structure type [tetragonal, P42/m, a = 6.8058(7) A, c = 9.7597(14) A, Z = 2], which consists of central uranium(IV) atoms coordinated by P2S64– anions (staggered conformation). The anions are two-dimensional connectors for four uranium cations arranged in one plane. Abstract Complexes of Co(II), Ni(II) and Cu(II) with the title ligand, L, have been prepared and characterized through elemental analysis, conductance measurements, electronic and infrared spectra, and magnetic measurements. For all hexafluorophosphate complexes monomeric species with CuL2 2+ and ML3 2+ (M=Co,Ni,Cu) have been isolated. In the oxalate complexes the C2O4 = group act as a bidentate bridge to form compounds with probable dimeric (copper(II)) or polymeric (cobalt(II) and nickel(II)) structures.

We synthesized a series of serum-stable covalently linked drugs derived from 3'-C-methyladenosine (3'-Me-Ado) and valproic acid (VPA), which are ribonucleotide reductase (RR) and histone deacetylase (HDAC) inhibitors, respectively. While the combination of free VPA and 3'-Me-Ado resulted in a clear synergistic apoptotic effect, the conjugates had lost their HDAC inhibitory effect as well as the corresponding apoptotic activity. Two of the analogs, 2',5'-bis-O-valproyl-3'-C-methyladenosine (A160) and 5'-O-valproyl-3'-C-methyladenosine (A167), showed promising cytotoxic activities against human hematological and solid cancer cell lines. Abstract A method is described for the rapid purification of radioactively labeled deoxyribonucleoside triphosphates from their spontaneously emerging hydrolysis products deoxyribonucleoside diphosphate, deoxyribonucleoside monophosphate, and deoxyribonucleoside. The separations which are finished within 3 min or less are carried out on a 0.1X5 cm column filled with LiChrosorb-NH2 , using isocratic elution with 0.025 м potassium phosphate, pH 6 .8 , in a high-pressure liquid chromatograph at room temperature and a flow rate of 30 ml · h-1(flow velocity 63.7 cm·min-1).

A fully automated method including column-switching and isocratic high-performance liquid chromatography (HPLC) was developed for quantitative analysis of the new antidepressant reboxetine, a noradrenaline reuptake inhibitor. After serum injection into the HPLC system and on-line sample clean-up on a silica C8 (10x4.0 mm I.D.) clean-up column with an eluent consisting of 2.5% acetonitrile in deionized water, the chromatographic separation was performed on an analytical column (Lichrospher CN; 250x4.6 mm I.D.) with an eluent of acetonitrile-aqueous potassium phosphate buffer (0.008 M, pH 6.4) (50:50).
