Water-soluble hyperbranched polyglycerol photosensitizer for enhanced photodynamic therapy

2020

Porphyrin and its derivatives as promising photosensitizers have been widely utilized in photodynamic therapy (PDT). However, the water-solubility and biocompatibility of porphyrins are the typical bottlenecks in clinical PDT processes. Herein, we successfully fabricated a new type of water-soluble, hyperbranched polyglycerol photosensitizer through one-step esterification between water-soluble hyperbranched polyglycerol (hbPG) and fluorophenylporphyrin (FP). Compared with the linear control polymer, i.e., FP covalently connected with linear polyethylene glycol (LPEG), the hyperbranched structures bearing multiple porphyrin units generated more singlet oxygen and exhibited higher cytotoxici…

Synergistic Anticancer Therapy by Ovalbumin Encapsulation-Enabled Tandem Reactive Oxygen Species Generation

2020

Abstract The anticancer efficacy of photodynamic therapy (PDT) is limited due to the hypoxic features of solid tumors. We report synergistic PDT/chemotherapy with integrated tandem Fenton reactions mediated by ovalbumin encapsulation for improved in vivo anticancer therapy via an enhanced reactive oxygen species (ROS) generation mechanism. O2 .− produced by the PDT is converted to H2O2 by superoxide dismutase, followed by the transformation of H2O2 to the highly toxic .OH via Fenton reactions by Fe2+ originating from the dissolution of co‐loaded Fe3O4 nanoparticles. The PDT process further facilitates the endosomal/lysosomal escape of the active agents and enhances their intracellular deliv…

The efficacy of photodynamic therapy in rat tongue dysplasia

2017

Background Photodynamic therapy (PDT) using 5-aminolevulinic acid (ALA) has previously shown promising results in cancerous cell destruction. The present study was conducted to evaluate the efficacy of this treatment option on oral epithelial dysplasia in Wistar rats. Furthermore, microscopic effects of systemic versus topical administration of ALA before laser illumination was assessed. Material and Methods Thirty male Wistar rats (200- 250 grams) were used in the present study. Tongue dysplasia was induced by a daily delivery of a 20 ppm solution of 4-nitroquinoline -1- oxide (4NQO) for 3 months. Then, rats were divided into 3 groups of 10 including, group 1 that was received systemic ALA…

Long-Segment Early Squamous Cell Carcinoma of the Proximal Esophagus: Curative Treatment and Long-Term Follow-Up after 5-Aminolevulinic Acid (5-ALA)-…

2006

Photodynamic therapy (PDT) is an established treatment for the ablation of dysplastic Barrett's epithelium and early esophageal carcinoma, but no data have been published on curative 5-aminolevulinic acid (5-ALA)-PDT for long-segment early esophageal cancer with infiltration of the upper sphincter. We describe successful curative treatment of an unusual early long-segment squamous cell carcinoma (uT1a,N0) of the proximal esophagus (18 - 28 cm aborally), with three sessions of 5-ALA-PDT. Endoscopic and endosonographic follow-up macroscopically showed a complete remission of the tumor, proven by biopsy showing total histological ablation. After a follow-up period of 23 months, there has been …

Photodynamic Therapy in Subfoveal Choroidal Neovascularisation

2002

Summary The exudative form of age-related macular degeneration (AMD) is the leading cause of blindness among people over 50 years of age in the western world. In 2–10% of cases a subfoveal choroidal neovascularisation develops. Conventional laser treatment leads to an unspecific destruction of the CNV including adjacent retinal structures. In addition, significant and immediate vision loss occurs, particularly in cases with central, subfoveal localization of the CNV.Photodynamic therapy (PDT) employing photochemical activation of verteporfin as a photosensitizer induces selective occlusion of CNV with minimal damage to the neurosensory retina and does not impair visual acuity. It became ava…

Focal Therapy for Prostate Cancer: Complications and Their Treatment

2021

Focal therapy is a modern alternative to selectively treat a specific part of the prostate harboring clinically significant disease while preserving the rest of the gland. The aim of this therapeutic approach is to retain the oncological benefit of active treatment and to minimize the side-effects of common radical treatments. The oncological effectiveness of focal therapy is yet to be proven in long-term robust trials. In contrast, the toxicity profile is well-established in randomized controlled trials and multiple robust prospective cohort studies. This narrative review summarizes the relevant evidence on complications and their management after focal therapy. When compared to whole glan…

Retrospective clinical evaluation of root canal treatment with or without photodynamic therapy for necrotic teeth and teeth subjected to retreatment.

2021

PURPOSE To compare the effect of photodynamic therapy (PDT) as a procedure to complement root canal treatment (RCT), for both primary treatment and retreatment. METHODS This was a retrospective study based on analysis of clinical records. A total of 214 teeth that had undergone RCT on either a primary or retreatment basis, with or without complementary PDT, were evaluated. For 118 teeth that met the previously established inclusion criteria, the time until healing was evaluated. Complementarily, the need for application of calcium hydroxide (CaHy) between visits and the number of visits necessary for completing the treatment were assessed. Data were analyzed using the Mann-Whitney and χ2 te…
