Stereoselective Synthesis of Enantiomerically Pure Nupharamine Alkaloids from Castoreum

An animalic note: The first total synthesis of the all-cis nupharamine 2, an alkaloid from beaver castoreum, is based on the stereoselective domino Mannich-Michael reaction of N-galactosylfurylaldimine to give 1 (Piv = pivaloyl), subsequent conjugate cuprate addition, and stereoselective protonation of the enolate. These reactions are all controlled by the carbohydrate. Protonation of the enolate after cleavage of the auxiliary leads to epimer 3.

Slow relaxation of magnetization in a {Fe6Dy} complex deriving from a family of highly symmetric metallacryptands

The synthesis and characterization of a new family of isostructural {Fe6Ln} complexes, (pipH)3{Fe6Ln(shiH)3(shi)6}·1.5 pip·xH2O (Ln = Gd (1), Dy (2), Tb (3) and Y (4), pip = piperidine, and shi3− = salicylhydroxamic acid) is reported. The resulting compounds possess an exceptionally unique structure of a metallacrown-like motif while the overall complexes feature more the structure of metallacryptates. Magnetic studies are reported and reveal that the {Fe6Dy} analogue belongs to the single-molecule magnet (SMM) family.

Bei der Thermolyse der Diazoverbindung 1 in Ethanol bzw. Piperidin oder Morpholin entstehen die Enole 3a–c von β-Ketocarbonsaurederivaten. Z- und E-Konfigurationen stehen miteinander im Gleichgewicht. Die Bestimmung der Geschwindigkeits-konstanten zeigt, das sich zunachst bevorzugt die Z-Formen bilden; im Gleichgewicht dominieren jedoch schlieslich in den Thermolysemedien die E-Isomeren. In aprotischen, apolaren oder masig polaren Medien uberwiegen dagegen die Z-Isomeren mit einer intramolekularen Wasserstoffbrucke. ROCK has been implicated in many diseases ranging from glaucoma to spinal cord injury and is therefore an important target for therapeutic intervention. In this study, we have designed a series of 1-(4-(1H-indazol-5-yl)piperazin-1-yl)-2-hydroxy(or 2-amino) analogs and a series of 1-(4-(1H-indazol-5-yl amino)piperidin-1-yl)-2-hydroxy(or 2-amino) inhibitors of ROCK-II. SR-1459 has IC50 = 13 nM versus ROCK-II while the IC50s for SR-715 and SR-899 are 80 nM and 100 nM, respectively. Many of these inhibitors, especially the 2-amino substituted analogs for both series, are modest/potent CYP3A4 inhibitors as well. The Peptide Hemopressin Acts through CB1Cannabinoid Receptors to Reduce Food Intake in Rats and Mice

Hemopressin is a short, nine amino acid peptide (H-Pro-Val-Asn-Phe-Lys-Leu-Leu-Ser-His-OH) isolated from rat brain that behaves as an inverse agonist at the cannabinoid receptor CB1, and is shown here to inhibit agonist-induced receptor internalization in a heterologous cell model. Since this peptide occurs naturally in the rodent brain, we determined its effect on appetite, an established central target of cannabinoid signaling. Hemopressin dose-dependently decreases night-time food intake in normal male rats and mice, as well as in obeseob/obmale mice, when administered centrally or systemically, without causing any obvious adverse side effects. A 2,3,4-tri-O-pivaloylated beta-D-galactopyranosyl azide bearing a hydroxy-functionalized spacer unit at the C-6 position of the galactose was synthesized and immobilized on the solid phase by using a polymer-bound chlorosilane. The azide was reduced to the corresponding galactopyranosylamine, which served as a versatile chiral auxiliary in highly diastereoselective Ugi four-component condensation reactions at ambient temperature. Background The endocannabinoid (eCB) 2-arachidonoylglycerol (2-AG) is a known modulator of inflammation and ligand to both, pro-inflammatory cannabinoid receptor 1 (CB1) and anti-inflammatory CB2. While the role of both receptors in atherogenesis has been studied extensively, the significance of 2-AG for atherogenesis is less well characterized.
