Cetuximab +/- chemotherapy enhances dendritic cell-mediated phagocytosis of colon cancer cells and ignites a highly efficient colon cancer antigen-sp…
2012
Cetuximab is a human/mouse chimeric IgG1 monoclonal antibody (mAb) to epidermal growth factor receptor, approved for colorectal carcinoma treatment in combination with chemotherapy. The immune-mediated effects elicited by its human fraction of crystallization moiety might critically contribute to the overall anti-tumor effectiveness of the antibody. We therefore investigated cetuximab ability to promote colon cancer cell opsonization and phagocytosis by human dendritic cells (DCs) that are subsequently engaged in antigen-cross presentation to cytotoxic T-lymphocyte (CTL) precursors. Human colon cancer cell lines were evaluated for susceptibility to DC-mediated phagocytosis before and after …
Immunomorphological Patterns of Chaperone System Components in Rare Thyroid Tumors with Promise as Biomarkers for Differential Diagnosis and Providin…
2023
Hurthle cell (HC), anaplastic (AC), and medullary (MC) carcinomas are low frequency thyroid tumors that pose several challenges for physicians and pathologists due to the scarcity of cases, information, and histopathological images, especially in the many areas around the world in which sophisticated molecular and genetic diagnostic facilities are unavailable. It is, therefore, cogent to provide tools for microscopists to achieve accurate diagnosis, such as histopathological images with reliable biomarkers, which can help them to reach a differential diagnosis. We are investigating whether components of the chaperone system (CS), such as the molecular chaperones, can be considered dependabl…
Hsp60 Protects against Amyloid β Oligomer Synaptic Toxicity via Modification of Toxic Oligomer Conformation
2019
Alzheimer's disease (AD) is the leading cause of dementia worldwide. While the etiology of AD remains uncertain, neurotoxic effects of amyloid beta oligomers (Aβo) on synaptic function, a well-established early event in AD, is an attractive area for the development of novel strategies to modify or cease the disease's progression. In this work, we tested the protective action of the mitochondrial chaperone Hsp60 against Aβo neurotoxicity, by determining the direct effect of Hsp60 in changing Aβo toxic conformations and thus reducing their dysfunctional synaptic binding and consequent suppression of long-term potentiation. Our data suggest that Hsp60 has a direct impact on Aβo, resulting in a…
Iron-Catalyzed C(sp2)–C(sp3) Cross-Coupling of Chlorobenzenesulfonamides with Alkyl Grignard Reagents: Entry to Alkylated Aromatics
2018
Alkylated benzosulfonamides are compounds of high importance in organic synthesis, including the production of pharmaceuticals, agrochemicals, and plasticizers. We report the iron-catalyzed C(sp2)–C(sp3) cross-coupling of chlorobenzosulfonamides with alkyl Grignard reagents under mild and sustainable conditions. Electronically and sterically varied benzosulfonamides as well as challenging alkyl organometallics containing β-hydrogen afford alkylated benzosulfonamides in high to excellent yields. Sulfonamide represents the most reactive activating group for iron-catalyzed cross-coupling. The process affords alkylated benzenesulfonamides poised for medicinal chemistry applications and traceles…
Effect of the orientation on the photooxidation behaviour of polymer films
2016
Abstract The effect of the orientation on the photooxidation of polymers is quite controversial. Both improvement of the photo-resistance along the oriented direction and worsening of the photooxidation kinetic have been reported. In this work the effect of the orientation has been investigated on films of PE, PP and PET with different degree of orientation and crystallinity. The results show that the photo-stability, measured from the decay of the elongation at break curve as a function of the irradiation time, improves in the more oriented direction, but this is more and more relevant with increasing the crystallinity degree. It has been hypothesized that the reinforcing action of the ori…
Fluorescence studies of hydrophobically-modified poly(N-isopropylacrylamides) or how to play accordion on a liposome
1991
The coil/globule transition of polymers in solution is a favourite topic of discussion among polymer theoreticians. Here, this phenomenon is used to play a “molecular accordion” built with a thermosensitive polymer anchored to liposomes, a tune which mimics a natural process occuring in cell membranes.
Spatial Demixing of Ring and Chain Polymers in Pressure-Driven Flow
2019
We investigate mixtures of ring and linear polymers in solution at various number ratios, ranging from pure chains to pure rings, and at densities around the overlap concentration. In bulk and at r...
Optical data storage in Langmuir-Blodgett-Kuhn multilayer assemblies of azo-dye side chain LC-polymers
1991
We describe the preparation of LB-films of novel polymers containing azobenzene dye side chains. X-ray refection measurements, surface-plasmon experiments were used to determine the structure of LB-films which had been transferred onto a substrate. Illumination with UV-light causes trans-cis isomerisation of the azobenzene side groups. This effect can be used for optical data storage and will be discussed in detail.
Cell adhesion on UV-crosslinked polyurethane gels with adjustable mechanical strength and thermoresponsiveness
2021
Temperature-responsive polyurethane (PU) hydrogels represent a versatile material platform for modern tissue engineering and biomedical applications. However, besides intrinsic advantages such as high mechanical strength and a hydrolysable backbone composition, plain PU materials are generally lacking bio-adhesive properties. To overcome this shortcoming, the authors focus on the synthesis of thermoresponsive PU hydrogels with variable mechanical and cell adhesive properties obtained from linear precursor PUs based on poly(ethylene glycol)s (pEG) with different molar masses, isophorone diisocyanate, and a dimerizable dimethylmaleimide (DMMI)-diol. The cloud point temperatures of the dilute,…
