Identification of Cysteine Residues in Human Cationic Amino Acid Transporter hCAT-2A That Are Targets for Inhibition by N-Ethylmaleimide

2013

In most cells, cationic amino acids such as l-arginine, l-lysine, and l-ornithine are transported by cationic (CAT) and y(+)L (y(+)LAT) amino acid transporters. In human erythrocytes, the cysteine-modifying agent N-ethylmaleimide (NEM) has been shown to inhibit system y(+) (most likely CAT-1), but not system y(+)L (Devés, R., Angelo, S., and Chávez, P. (1993) J. Physiol. 468, 753-766). We thus wondered if sensitivity to NEM distinguishes generally all CAT and y(+)LAT isoforms. Transport assays in Xenopus laevis oocytes established that indeed all human CATs (including the low affinity hCAT-2A), but neither y(+)LAT isoform, are inhibited by NEM. hCAT-2A inhibition was not due to reduced tran…

Real-Time Assembly Support System with Hidden Markov Model and Hybrid Extensions

2022

This paper presents a context-aware adaptive assembly assistance system meant to support factory workers by embedding predictive capabilities. The research is focused on the predictor which suggests the next assembly step. Hidden Markov models are analyzed for this purpose. Several prediction methods have been previously evaluated and the prediction by partial matching, which was the most efficient, is considered in this work as a component of a hybrid model together with an optimally configured hidden Markov model. The experimental results show that the hidden Markov model is a viable choice to predict the next assembly step, whereas the hybrid predictor is even better, outperforming in so…

Human cationic amino acid transporter gene hCAT-2 is assigned to 8p22 but is not the causative gene in lysinuric protein intolerance

1997

Lysinuric protein intolerance (LPI) is a recessively inherited amino acid disorder characterized by defective efflux of cationic amino acids at the basolateral membrane of the intestinal and renal tubular epithelium. Recently, cDNAs encoding the related proteins hCAT-2A and hCAT-2B have been cloned. These two carrier proteins are most likely the product of the same gene, hCAT-2. Using the hCAT-2B cDNA, we assigned the hCAT-2 gene to chromosome 8p22. Furthermore, by linkage analysis in Finnish LPI families, we ruled out that hCAT-2B is involved in LPI disease.

COVID-Guide: An app for Covid-19 triage and self-assessment

2021

Introduction. The unprecedented covid-19 pandemic has shown the weaknesses of health systems and opened new spaces for e-health and telemedicine. Recent literature states that chatbots, if implemented effectively, could be useful tools for quickly sharing information, promoting healthy behaviors, and helping reduce the psychological burden of isolation. The aim of this project is to develop and test a secure and reliable computerized decision support system (CDSS) in web-app and evaluate its use, usability and its outputs in a pre-specified way. Methods. A multidisciplinary team was recruited to plan and design, based on the SMASS medical CDSS, the scenarios of the COVID-Guide web-app, a se…

Randomized pilot study and qualitative evaluation of a clinical decision support system for brain tumour diagnosis based on SV 1H MRS: Evaluation as …

2014

The results of a randomized pilot study and qualitative evaluation of the clinical decision support system Curiam BT are reported. We evaluated the system's feasibility and potential value as a radiological information procedure complementary to magnetic resonance (MR) imaging to assist novice radiologists in diagnosing brain tumours using MR spectroscopy (1.5 and 3.0T). Fifty-five cases were analysed at three hospitals according to four non-exclusive diagnostic questions. Our results show that Curiam BT improved the diagnostic accuracy in all the four questions. Additionally, we discuss the findings of the users' feedback about the system, and the further work to optimize it for real envir…

Internet based exercise management system for worksite exercise and well-being intervention

2012

Reijo Kangas. 2012. Internet based exercise management system for worksite exercise and well-being intervention. University of Jyväskylä. Department of Sport Sciences, Master’s thesis in Sport and Exercise Psychology, 105 pages. The studies of exercise motivation theories can be used for the requirements of exercise management software systems. User-friendly mobile and web applications may wake up people’s interest of their own awareness of unhealthy manners like obesity, sedentary behavior and possible mental health problems of inactivity. The aim of the present study was to plan, organize and evaluate an internet based exercise intervention for workers. Other purpose of the study was to o…

Role of Neutral Amino Acid Transport and Protein Breakdown for Substrate Supply of Nitric Oxide Synthase in Human Endothelial Cells

2003

Endothelial dysfunction is often associated with a relative substrate deficiency of the endothelial nitric oxide synthase (eNOS) in spite of apparently high intracellular arginine concentrations. For a better understanding of the underlying pathophysiological mechanisms, we aimed to characterize the intracellular arginine sources of eNOS. Our previous studies in human endothelial EA.hy926 cells suggested the existence of two arginine pools: pool I can be depleted by extracellular lysine, whereas pool II is not freely exchangeable with the extracellular space, but accessible to eNOS. In this study, we demonstrate that the eNOS accessible pool II is also present in human umbilical vein endoth…

Les dispositifs de prise en charge de la difficulté scolaire à l’école élémentaire : étude de leur fonctionnement et de leurs effets sur la réussite …

2018

With the Jules Ferry laws ensuring free and compulsory schooling, equal chances of succeeding were expected in a meritocratic established school system. Reality proved otherwise: attending school still does not allow all schoolchildren to progress in the same way, as some of them encounter difficulties from the beginning. The struggle against academic failure cannot stop a considerable amount of pupils from finishing school without satisfactorily mastering the basic elements of the programmes. Moreover, recent research works have seemed to prove programmes providing assistance for underachievers to be ineffective. This thesis offers a comparative evaluation of such support systems in element…

MODULAR KNOWLEDGE REPRESENTATION IN ADVISOR AGENTS FOR SITUATION AWARENESS

2011

A modular knowledge representation framework for conversational agents is presented. The approach has been realized to suit the situation awareness paradigm. The modularity of the framework makes possible the composition of specific modules that deal with particular features, simplifying both the chatbot design process and its smartness. As a proof of concepts we have developed a modular, situation awareness oriented, KB for a conversational agent, which plays the role of an advisor aimed at helping a user to be in charge of a virtual town, inspired to the SimCity series game. The agent makes an extensive use of semantic computing techniques and is able to perceive, comprehend and project c…
