Abstract Temsirolimus (Torisel®) is a specific inhibitor of the mTOR kinase with antitumor activity in patients with relapsed or refractory mantle cell lymphoma. In a phase 3, randomized, open-label study, patients treated with temsirolimus 175 mg weekly 3 times followed by 75 mg weekly (175/75-mg) had significantly longer progression-free survival (PFS) than those treated with investigator’s choice therapy (p-value temsirolimus: investigator’s choice = 0.0009; hazard ratio = 0.44; 97.5% CI = 0.25, 0.78; Hess et al. Chiral bis(amino alcohol)oxalamides as ligands for asymmetric catalysis. Ti(IV) catalyzed enantioselective addition of diethylzinc to aldehydes

Abstract Several chiral bis(aminoalcohol)oxalamides with C 2 -symmetry have been prepared and used as ligands for the enantioselective addition of diethylzinc to aromatic and aliphatic aldehydes. The reaction proceeds in the presence of titanium isopropoxide to give the corresponding ( S )-alcohols with ee up to 78%. In the absence of Ti(IV), the alcohols with the opposite configuration are obtained.

Has pharmaceutical research become more scientific?

Pharmaceutical research continues to be highly empirical, despite some recent writings arguing that it has become more scientific. Contrary to a study asserting that Mevacor, Tagamet, and Capoten demonstrate the success of targeted research, these drugs were discovered only after many years of following leads. Two hypotheses implied by the proposition that pharmaceutical research has become more scientific are tested. The first hypothesis is that the uncertainty of research has fallen. The second is that the number of publications by scientists in the industry has increased. Both are rejected.

In a prospective study conducted since 1983, the hormone-receptor status of primary breast carcinomas was investigated using immunohistochemical (ER-ICA, PgR-ICA) and biochemical (DCC) methods. The degree of immunohistochemical staining was evaluated according to the immunoreactive score (IRS) devised by Remmele and Stegner [Frauenarzt 28, 41-43 (1987)]. The findings obtained using the biochemical radioactive-ligand-binding assay (cutoff level, 20 fmol/mg) and those obtained using qualitative immunohistochemical methods were in agreement in 72.5% (ER-ICA) and 72.2% (PgR-ICA) of cases.
