Synthesis and biological evaluation of a D-ring-contracted analogue of lamellarin D

2017

A D-ring contracted analogue of the strongly cytotoxic marine pyrrole alkaloid lamellarin D was synthesized and investigated for its antiproliferative action towards a wild type and a multidrug resistant (MDR) cancer cell line. The compound was found to inhibit tumor cell growth at submicromolar concentrations and showed a lower relative resistance in the MDR cell line than the antitumor drug camptothecin to which lamellarin D shows cross resistance and with which lamellarin D shares the same binding site.

Identity Double-Proton Transfer in (3Z)-3-Hydroxy-1,4-di(quinolin-2-yl)but-3-en-2-one

Although there is a very fast (on the NMR timescale) double-proton transfer in (1Z,3Z)-3-hydroxy-4-quinolin-2-yl-1-quinolin-2(1H)-ylidenbut-3-en-2-one (the product of the condensation of ethyl oxalate with 2-lithiomethylquinoline), it is the only species present in chloroform solution. Comparison of the product of condensation of ethyl oxalate with 2-lithiomethyl derivatives of pyridine (recent studies) and quinoline (present studies) shows that benzoannulation considerably affects the tautomeric equilibrium. One-dimensional oxalato-bridged metal(II) compounds of formula [M(-ox)(L)2]n [L = 3-hydroxypyridine (pyOH) or isoquinoline (isq)] have been synthesised and characterised by FT-IR spectroscopy, TG-DTA techniques, variable-temperature magnetic measurements and X-ray diffraction methods. The complexes [M(-ox)(pyOH)2]n [M= Co (1), Ni (2)] are isomorphous and crystallise in the orthorhombic space group Pnab. The compounds [M(-ox)(isq)2]n [M= Co (3), Ni (4), Cu (5)] are also isomorphous and belong to the monoclinic space group C2/c. Superparamagnetic iron oxide/oleic acid nanoparticles bearing lipid-like organosilicon N-(2-hydroxyethyl)-1,2,3,4-tetrahydroisoquinoline derivatives have been synthesized with the aim of their potential biomedical application. X-ray diffraction analysis, Dynamic light-scattering measurements, method of magnetogranulometry and some others have been employed to investigate the morphology and properties of the nanoparticles synthesized. An Alternative Pathway of Imiquimod-Induced Psoriasis-Like Skin Inflammation in the Absence of Interleukin-17 Receptor A Signaling

Topical application of imiquimod (IMQ) on the skin of mice induces inflammation with common features found in psoriatic skin. Recently, it was postulated that IL-17 has an important role both in psoriasis and in the IMQ model. To further investigate the impact of IL-17RA signaling in psoriasis, we generated IL-17 receptor A (IL-17RA)-deficient mice (IL-17RA(del)) and challenged these mice with IMQ. Interestingly, the disease was only partially reduced and delayed but not abolished when compared with controls. Tautomeric preferences of phthalones and related compounds

Abstract Multinuclear magnetic resonance and IR spectra prove that although 2-(diacylmethyl)pyridines and 2-(diacylmethyl)quinolines are β-diketones, their proton transfer product present in chloroform solution is not ketoenol but enaminone (earlier opinions were contradictory). Quinoline derivatives are less zwitterionic by character than the respective pyridyl congeners. The β-diketone form itself may also be rarely present in the solution. X-ray data show that 2-(2(1H)-pyridinylidene)-1H-indene-1,3(2H)-dione, i.e., enaminone tautomer of 2-(pyridin-2-yl)-2H-indene-1,3-dione, is also the only form present in crystal. Abstract The Congolese liana Ancistrocladus likoko (Ancistrocladaceae) produces naphthylisoquinoline alkaloids that are, chemotaxonomically remarkable, all based on the same coupling type, with the biaryl axis located between C-5 and C-8′. A unique series of six biaryl natural products displaying four different coupling types (5,1′, 7,1′, 7,8′, and 5,8′) were isolated from the roots of the West African liana Ancistrocladus abbreviatus (Ancistrocladaceae). Although at first sight structurally diverse, these secondary metabolites all have in common that they belong to the rare group of naphthylisoquinoline alkaloids with a fully dehydrogenated isoquinoline portion.
