Non-uniformity of displacement and strain within the Achilles tendon is affected by joint angle configuration and differential muscle loading.

Although the Achilles tendon (AT) has been studied for more than a century, a complete understanding of the mechanical and functional consequences of AT structural organization is currently lacking. The aim of this study was to assess how joint angle configuration affects subtendon displacement and strain of soleus (SOL) and lateral gastrocnemius (LG) muscles. Knots sutured onto SOL and LG subtendons of 12 Wistar rats, were videotaped to quantify displacements and the ankle torque was assessed for different isometric activation conditions (i.e., individual and simultaneous) of the triceps surae muscles. Fibril formation and toxicity of the non-amyloidogenic rat amylin peptide.

Full-length native rat amylin 1-37 has previously been widely shown to be unable to form fibrils and to lack the toxicity of the human amylin form leading to its use as a non-amyloidogenic control peptide. A recent study has suggested that rat amylin 1-37 forms amyloidogenic β-sheet structures in the presence of the human amylin form and suggested that this property could promote toxicity. Using TEM analysis we show here fibril formation by synthetic rat amylin 1-37 and 8-37 peptides when the lyophilized HPLC purified peptides are initially dissolved in 20 mM Tris-HCl. Dissolution of synthetic rat amylin 1-37 and 8-37 peptides in H(2)O or phosphate buffered saline failed to produce fibrils.…

Nuclear calcium signaling by inositol trisphosphate in GH3 pituitary cells

It has been proposed that nuclear and cytosolic Ca2+ ([Ca2+]N and [Ca2+]C) may be regulated independently. We address here the issue of whether inositol trisphosphate (IP3) can, bypassing changes of [Ca2+]C, produce direct release of Ca2+ into the nucleoplasm. We have used targeted aequorins to selectively measure and compare the changes in [Ca2+]C and [Ca2+]N induced by IP3 in GH3 pituitary cells. Heparin, an IP3 inhibitor that does not permeate the nuclear pores, abolished the [Ca2+]C peaks but inhibited only partly the [Ca2+]N peaks. One autapomorphic character restricted to all Metazoa including Porifera [sponges] is the existence of transmembrane receptor tyrosine kinases (RTKs). In this study we screened for molecules from one subfamily within the superfamily of the insulin receptors. AbstractThe pituitary adenylate cyclase activating polypeptide (PACAP) type I receptor belongs to the glucagon/secretin/vasoactive intestinal polypeptide (VIP) receptor family. We mutated and deleted an amino acid residue (E261) which is located within the second intracellular loop of the rat PACAP type I receptor and which is highly conserved among the receptor family.
