The blood-brain barrier prevents the passage of toxic compounds from blood circulation into brain tissue. Unfortunately, drugs for the treatment of neurodegenerative diseases, brain tumors, and other diseases also do not cross the blood-brain barrier. In the present investigation, we used isolated porcine brain capillary endothelial cells and a flow cytometric calcein-AM assay to analyze inhibition of P-glycoprotein, a major constituent of the blood-brain barrier. We tested 8 alkamides isolated from Echinacea angustifolia and found that four of them inhibited P-glycoprotein-mediated calcein transport in porcine brain capillary endothelial cells.

Size-consistent ab initio calculation of the electric quadrupole moment of Cl2

Abstract The molecular electric quadrupole moment ( Θ ) of Cl 2 has been calculated using SDCI, and (SC) 2 -SDCI wave functions as well as CCSD, CCSD(T), and CC3 methods. All these correlation methods are single reference. All of them, but SDCI, are free of the size-extensivity error. The variation of Θ from the separated atoms to the equilibrium region is reported. The present results leads to an estimated value of 2.3520 a.u. (10.55 × 10 −40 Cm 2 ) corresponding to a CC(3) calculation at the CBS approach and including the ro-vibrational and thermal averaging corrections. This value is compatible with two experimental values and points to one of them as slightly more reliable.

In the literature on Kant’s philosophy, it is almost universally ignored that Baumgarten used the notion of positing in a technical sense before Kant. In this article, I try to fill this gap in the literature by providing an analysis of Kant’s notion of positing in relation to Baumgarten’s one. I first show how Kant's differentiation of relative and absolute positing is not simply an alternative to Baumgarten's notion of existence, but is instead an alternative to his (usually ignored) notion of positing; I then develop a positive account of Kant’s notion of positing.
Vertical spectrum of ethene: uncontracted versus contracted correlation methods and the role of the adapted molecular orbitals

Abstract Ethylene vertical excitation energies (VEE) below 9.5 eV have been calculated with the complete active-space singles and doubles configuration interaction self-consistent size-consistent dressing (SC) 2 CAS–SDCI. The mean of the absolute deviation from experiment for the calculated VEE are 0.14, 0.05 and 0.03 eV for three different molecular orbital (MO) sets tried. The results show that an uncontracted method and a realistic MO set avoid the difficulties present in the calculation of the strongly mixed Rydberg-valence 1B 1u states.
Estimation of ADME Properties in Drug Discovery: Predicting Caco-2 Cell Permeability Using Atom-Based Stochastic and Non-stochastic Linear Indices

The in vitro determination of the permeability through cultured Caco-2 cells is the most often-used in vitro model for drug absorption. In this report, we use the largest data set of measured P(Caco-2), consisting of 157 structurally diverse compounds. Linear discriminant analysis (LDA) was used to obtain quantitative models that discriminate higher absorption compounds from those with moderate-poorer absorption. The best LDA model has an accuracy of 90.58% and 84.21% for training and test set. The percentage of good correlation, in the virtual screening of 241 drugs with the reported values of the percentage of human intestinal absorption (HIA), was greater than 81%. Goal pursuit may affect older people’s outdoor mobility, a correlate of quality of life. This study investigated associations of flexible and tenacious goal pursuit with perceived autonomy in participation outdoors and life-space mobility. Participants were community-dwelling older people aged 79–93 years (n=185). Tenacious goal pursuit (TGP) and flexible goal adjustment (FGA) were self-reported with separate scales (ranges 5–25). The need to grow, learn and develop – how does management affect motivation for professional development?

This article argues that knowledge management and social recognition is important for organisational learning and professional self-esteem in academic libraries. An anonymous survey was issued in 2016 to investigate how library staff’s self-esteem is affected by how they experience their management’s view and overview of their knowledge. The need for what Axel Honneth refers to as social recognition will also be discussed as an important part of how professional self-esteem and work satisfaction is experienced and further how this affects motivation to participate in professional development.
