Delivery of proteins into living cells by reversible membrane permeabilization with streptolysin-O

The pore-forming toxin streptolysin O (SLO) can be used to reversibly permeabilize adherent and nonadherent cells, allowing delivery of molecules with up to 100 kDa mass to the cytosol. Using FITC-labeled albumin, 10 5 –10 6 molecules were estimated to be entrapped per cell. Repair of toxin lesions depended on Ca 2+ -calmodulin and on intact microtubules, but was not sensitive to actin disruption or to inhibition of protein synthesis. Metabotropic glutamate receptors activate phospholipase D in astrocytes through a protein kinase C-dependent and Rho-independent pathway.

Metabotropic glutamate receptors (mGluRs) are G protein-coupled receptors that mediate phospholipase D (PLD) activation in brain, but the mechanism underlying this response remains unclear. Here we used primary cultures of astrocytes as a cell model to explore the mechanism that links mGluRs to PLD. Glutamate activated both phospholipase C (PLC) and PLD with equal potency and this effect was mimicked by L-cysteinesulfinic acid, a putative neurotransmitter previously shown to activate mGluRs coupled to PLD, but not PLC, in adult brain. This contribution deals with the centrality of family relationships for the production of individual well-being, both on a cognitive level, i.e. as an influence on satisfaction with one’s own life and, therefore, as the outcome of a cognitive evaluation, and on an experiential and emotional level, i.e. as an influence on moment-to-moment experience. It starts from an analysis of subjective well-being, arriving at a concept of relational well-being, whilst proposing an approach of rhythmanalysis as a tool for understanding the changes occurring in family dynamics in the post-Covid era.

It first reviews this concept and draws attention to the multidimensionality of the construct, considering the underlying cognitive, emotional, and behavioral dimensions of consumer engagement. A book of the Indian spiritual master Jddu Krishnamurti (Education and the Significance of life), translated in 1958 by Aldo Visalberghi and published by La Nuova Italia, contained anti-educational and provocative proposals unrelated to the interests of the pedagogists. There was, by the academic world, a timid curiosity about the critical and polemical elements of the Krishnamurtian philosophy, that ignored or misunderstood the new and original conception of the school, that pedagogy can now reconsiders for a critic reflection about the new educational emergencies.

The school-based promotion of physical activity (PA) is a great opportunity to reach the majority of school-aged children. Aside from many physical health benefits, participation in physical activities can foster social well-being and interaction. The purpose of this study was to investigate changes in PA and school-related social factors, as well as their associations in school-aged children in schools which participated in the Finnish Schools on the Move programme and its pilot phase in 2010–2012.
