Metal stresses modify soluble proteomes and toxin profiles in two Mediterranean strains of the distributed dinoflagellate Alexandrium pacificum

WOS:000789651000009; HABs involving Alexandrium pacificum have been reported in metal-contaminated ecosystems, suggesting that this distributed species adapts to and/or can tolerate the effects of metals. Modifications in soluble proteomes and PST contents were characterized in two Mediterranean A. pacificum strains exposed to mono- or polymetallic stresses (zinc, lead, copper, cadmium). These strains were isolated from two anthropized locations: Santa Giusta Lagoon (Italy, SG C10-3) and the Tarragona seaport (Spain, TAR C5-4F). We have studied the dependence of the chemical reaction mechanism of L-lactate dehydrogenase (LDH) on the protonation state of titratable residues and on the level of the quantum mechanical (QM) description by means of hybrid quantum-mechanical/molecular-mechanical (QM/MM) methods; this methodology has allowed clarification of the timing of the hydride transfer and proton transfer components that hitherto had not been possible to state definitively. The general Stokes’s theorem gives a relationship between the integral of ω over the boundary of M and the integral of the exterior differential dω over M. Kresoxim-methyl is nowadays widely used to combat a diversity of common diseases affecting high-value crops. In this article, we report the development and characterization of two novel immunoassays for the analysis of this pioneer strobilurin fungicide, and for the first time, a validation study with food samples was performed. A direct and an indirect competitive immunoassay based on a new anti-kresoxim-methyl monoclonal antibody were developed for sensitive and specific chemical analysis. Optimized assays showed limits of detection of 0.1 μg/L. Abstract Residues of benzoylphenylurea insecticides (diflubenzuron, hexaflumuron, and flufenuxuron), carboxamide acaricides (hexythiazox), and carbamate insecticides (benfuracarb) were determined in 150 orange fruit samples from September 1998 to June 1999, to estimate exposure of the Valencian population to oranges contaminated with these newly developed pesticides. A multiresidue method based on matrix solid-phase dispersion (MSPD) microextraction was studied to determine the carbamate, benfuracarb, and urea insecticides, diflubenzuron, flufenoxuron hexaflumuron and hexythiazox, used in control of citrus pests. Optimisation of different parameters, such as the type of solid support for matrix dispersion, elution solvents and the clean-up step were carried out. The method used 0.5 g of orange sample, C8 bonded silica as MSPD sorbent and dichloromethane as eluting solvent. Perthamides C-F, potent human antipsoriatic cyclopeptides

Abstract Two new cyclopeptides, perthamides E and F were isolated from the polar extracts of the sponge Theonella swinhoei . The new structures, featuring an unprecedented β-amino acid unit (AHMOA), were determined by interpretation of NMR and MS data. The absolute configuration of the AHMOA residue was proposed on the basis of quantum chemical calculation of NMR chemical shifts. Perthamides were proved to inhibit TNF-α and IL-8 release in primary human keratinocytes cells and therefore could represent potentially leads for the treatment of psoriasis.
Lewis acid induced [4+3] cycloadditions of 2-silyloxyacroleins. Insights on the mechanism from a DFT analysis

The mechanism for the Lewis acid induced [4+3] cycloadditions of 2-(trimethylsilyloxy)acrolein with furan has been examined here through DFT calculations at B3LYP/6-31G* level. The mechanism is a three-step process initialized by the nucleophilic attack of furan to the β-conjugated position of acrolein yielding a zwitterionic intermediate. The key step on the formation of the seven-membered ring is the electrophilic attack of the furan residue to the carbonyl carbon in this intermediate. The endo selectivity experimentally observed is reproduced by the calculations.
An evaluation of a model ecosystem with DDT

The reproducibility, variability, and reliability of a simple aquatic-terrtestrial model ecosystem experiment was tested with p,p'-DDT. The variation among the model replicates as well as within the units was studied by using hierarchal analysis of variance. the complete budget of the chemical was calculated and a theoretical transport model was constructed. The degradation and accumulation of DDT was followed by gas chromatographic residue analysis, which showed good reproducibility of the experimental design.
