Abstract Background In eukaryotic cells, proteins are translocated across the ER membrane through a continuous ribosome-translocon channel. It is unclear to what extent proteins can fold already within the ribosome-translocon channel, and previous studies suggest that only a limited degree of folding (such as the formation of isolated α-helices) may be possible within the ribosome. Translational adaptation to heat stress is mediated by RNA 5‐methylcytosine in Caenorhabditis elegans

Abstract Methylation of carbon‐5 of cytosines (m5C) is a post‐transcriptional nucleotide modification of RNA found in all kingdoms of life. While individual m5C‐methyltransferases have been studied, the impact of the global cytosine‐5 methylome on development, homeostasis and stress remains unknown. Here, using Caenorhabditis elegans, we generated the first organism devoid of m5C in RNA, demonstrating that this modification is non‐essential. Using this genetic tool, we determine the localisation and enzymatic specificity of m5C sites in the RNome in vivo. Evidence for a novel cytoplasmic processing event in ribosome maturation in the sea urchin Paracentrotus lividus

In this work, we demonstrate the existence of a cytoplasmic processing step, never before described, involving both the pre-ribosomal subunits in the sea urchin Paracentrotus lividus. Northern-blot hybridization, primer extension, S1 mapping experiments and in situ hybridizations allowed us to demonstrate that cytoplasmic processed particles are successively re-imported into the nucleus, where maturation of their RNAs is completed prior to being exported to the cytoplasm. Our findings lead to the proposal of a new model of ribosome maturation and shuttling. Proteins from rat liver cytosol which stimulate mRNA transport. Purification and interactions with the nuclear envelope mRNA translocation system.

Two polysome-associated proteins with particular affinities for poly(A) have been purified from rat liver. These proteins stimulate the efflux of mRNA from isolated nuclei in conditions under which such efflux closely stimulates mRNA transport in vivo, and they are therefore considered as mRNA-transport-stimulatory proteins. Their interaction with the mRNA-translocation system in isolated nuclear envelopes has been studied. The results are generally consistent with the most recently proposed kinetic model of mRNA translocation. One protein, P58, has not been described previously. Electron microscopic study on the larval and adult corpus allatum of Oncopeltus fasciatus dallas (insecta, heteroptera)

1. The ultrastructure of the corpora allata of last larval instars and adults of Oncopeltus was studied. The unpaired gland undergoes submicroscopic alterations and shows signs of degradation in old animals. The organ is partly covered and penetrated by corpus cardiacum tissue. Axons with different types of neurosecretory granules form synaptoid contacts with the corpus allatum cells.

Ultrastructure of differentiating hemocytes in the embryo of Oncopeltus fasciatus dallas (insecta, heteroptera).

The hemocytes of Oncopeltus differentiate rather early during embryogenesis. They are segregated by the mesoderm soon after its formation (about 50h after egg deposition). Newly segregated hemocytes show the “typical” features of “embryonic” cells: many free ribosomes, a few strands of rough ER, the cisternae of which are considerably distended, electron lucent vacuoles around the periphery, and glycogen deposits. A few hours thereafter the hemocytes undergo striking subcellular changes. First, glycogen, electron lucent vacuoles and rough ER disappear and phagocytotic activity can be observed. Messenger RNA of the large subunit of ribulose-1,5-bisphosphate carboxylase from Chlamydomonas reinhardi. Isolation and properties.

Polysomes specifically synthesizing the large subunit of ribulose-1,5-bisphosphate carboxylase were isolated from Chlamydomonas reinhardi cells by the indirect immunoprecipitation method. Electrophoretic analysis showed that the immunoprecipitated polysomes were of chloroplast origin. The mRNA coding for the large subunit which was purified from immunoprecipitated polysomes migrated at the 19-S position on sucrose density gradients, and its molecular weight was estimated to be 7.3 x 10(5) by acid-urea/agarose gel electrophoresis. The mRNA was translated in vivo with a cell-free protein-synthesizing system derived from Escherichia coli to give full-length large-subunit polypeptides.

Peptide Bond Formation Mechanism Catalyzed by Ribosome

In this paper we present a study of the peptide bond formation reaction catalyzed by ribosome. Different mechanistic proposals have been explored by means of Free Energy Perturbation methods within hybrid QM/MM potentials, where the chemical system has been described by the M06-2X functional and the environment by means of the AMBER force field. According to our results, the most favorable mechanism in the ribosome would proceed through an eight-membered ring transition state, involving a proton shuttle mechanism through the hydroxyl group of the sugar and a water molecule.
