We have tested whether tetracyclines (TETs) are able to protect mice from lipopolysaccharide (LPS)-induced shock, a cytokine-mediated inflammatory reaction. Mice, injected with a single dose of tetracycline base (TETb; 1.5, 10 and 20 mg/kg of body weight) or doxycycline (DOXY; 1.5 mg/kg), were significantly protected from a lethal intraperitoneal injection of LPS (500 micrograms per mouse). TETs acted in early events triggered in response to LSP; in fact, they were no longer significantly protective if injected more than 1 h after the injection of endotoxin. A rat colon cancer cell line (CC531) was implanted into the livers of 85 WAG rats and in animals surviving the implantation the liver tumour was subsequently treated with sham laparotomy (n = 15), resection (n = 20), cryotherapy (n = 19) or laser therapy (n = 20). Local recurrence or persistence of the tumour in animals surviving the procedure was observed in 15 of 15 following sham laparotomy, in 0 of 15 following resection, in 3 of 17 following cryotherapy and in 10 of 14 following laser therapy. However, peritoneal disease and lung metastases were more frequent and extensive following resection as compared to cryotherapy or laser therapy.

Intestinal ischaemia is a major complication of shock syndromes causing translocation of bacteria and endotoxins and multiple organ failure in intensive care patients. The present study was designed to use microdialysis as a tool to monitor intestinal ischaemia after cardiac arrest and resuscitation in pigs. The influence of a guinea pig lymphokine preparation on the oxidative metabolism of human and guinea pig granulocytes of various sources was investigated. A dose-dependent increase of the oxidative burst following lymphokine challenge was observed. It occurred in unstimulated guinea pig peripheral polymorphonuclear leukocytes (PMN) and in prestimulated PMN obtained from the peritoneal cavity after glycogen injection as well. The lymphokine effect on the oxidative metabolism is not species-restricted because the guinea pig lymphokine preparation elicits an oxidative burst in human PMN, too.
