Regulatory T Cells and IL-10 Independently Counterregulate Cytotoxic T Lymphocyte Responses Induced by Transcutaneous Immunization

2011

Background: The imidazoquinoline derivate imiquimod induces inflammatory responses and protection against transplanted tumors when applied to the skin in combination with a cognate peptide epitope (transcutaneous immunization, TCI). Here we investigated the role of regulatory T cells (Treg) and the suppressive cytokine IL-10 in restricting TCI-induced cytotoxic T lymphocyte (CTL) responses. Methodology/Principal Findings: TCI was performed with an ointment containing the TLR7 agonist imiquimod and a CTL epitope was applied to the depilated back skin of C57BL/6 mice. Using specific antibodies and FoxP3-diphteria toxin receptor transgenic (DEREG) mice, we interrogated inhibiting factors after…

Spatial coherence and potential predictability of intraseasonal descriptors of the rainy season in Soudano-Sahelian Africa : application to the pearl…

2010

The aim of this thesis is twofold : (i) fill a lack of knowledge about the spatial coherence and seasonal predictability of the intraseasonal characteristics (ISC) of the soudano-sahelian rainy season. These ISC are the seasonal rainfall amount (S), the daily rainfall frequency > 1 mm (O), the daily rainfall mean intensity (I), the dates of the onset and withdrawal of the rainy season, the mean length of the dry (LDS) and/or wet spells (LWS) ; (ii) document the climate-agriculture relationship over the Niamey area based on the millet crop example. From FRIEND-AOC daily rainfall records (1950-2000) for a 136 stations-network located in Senegal, Mali, Burkina-Faso and Niger, the spatial cohere…

Principle considerations for the use of transcriptomics in doping research

2011

Over the course of the past decade, technical progress has enabled scientists to investigate genome-wide RNA ex- pression using microarray platforms. This transcriptomic approach represents a promising tool for the discovery of basic gene expression patterns and for identification of cellular signalling pathways under various conditions. Since doping substances have been shown to influence mRNA expression, it has been suggested that these changes can be detected by screening the blood transcriptome. In this review, we critically discuss the potential but also the pitfalls of this application as a tool in doping research. Transcriptomic approaches were considered to potentially provide resea…

Small genomes and the difficulty to define minimal translation and metabolic machineries

2015

The notion of minimal life has sparked the interest of scientists in different fields, ranging from the origin-of-life research to biotechnology-oriented synthetic biology. Whether the interest is focused on the emergence of protocells out of prebiotic systems or the design of a cell chassis ready to incorporate new devices and functions, proposing minimal combinations of genes for life is not a trivial task. Using comparative genomics and biochemistry of endosymbionts (i.e., intracellular mutualistic symbionts) and intracellular parasites, a decade ago we proposed the core of a minimal gene set for a simple heterotrophic cell adapted to a chemically complex environment. In this work, we di…

Concept and Development of Modular VLIW Processor Based on FPGA

2010

Modern FPGA chips, with their larger memory capacity and reconfigurability potential, are opening new frontiers in rapid prototyping of embedded systems. With the advent of high density FPGAs it is now possible to implement a high performance VLIW processor core in an FPGA. Architecture based on Very Long Instruction Word (VLIW) processors are an optimal choice in the attempt to obtain high performance level in embedded system. In VLIW architecture, the effectiveness of these processors depends on the ability of compilers to provide sufficient instruction level parallelism(ILP) in program code. Using advanced compiler technology could take these functions, This paper describes research resu…

Reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) as a user-friendly system to detect SARS-CoV-2 infection: a multicentric study

2021

Although reverse transcriptase quantitative PCR remains the gold standard to perform viral detection, reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) is already used to perform diagnosis of various infections. This work reports the results of a multicentric study performed in Sicily to evaluate the diagnostic power of an RT-LAMP kit for the diagnosis of SARS-CoV-2 infection on a total of 551 samples collected in January and February 2021, revealing sensitivity, specificity, accuracy, positive and negative predictive values ≥95%. Our results suggest the potential employment of this kit as a screening test to be used where fast and reliable results are demanded without …

The gut microbiota instructs the hepatic endothelial cell transcriptome

2021

Summary The gut microbiota affects remote organ functions but its impact on organotypic endothelial cell (EC) transcriptomes remains unexplored. The liver endothelium encounters microbiota-derived signals and metabolites via the portal circulation. To pinpoint how gut commensals affect the hepatic sinusoidal endothelium, a magnetic cell sorting protocol, combined with fluorescence-activated cell sorting, was used to isolate hepatic sinusoidal ECs from germ-free (GF) and conventionally raised (CONV-R) mice for transcriptome analysis by RNA sequencing. This resulted in a comprehensive map of microbiota-regulated hepatic EC-specific transcriptome profiles. Gene Ontology analysis revealed that …
