The extracorporeal perfusion of the female pig detrusor as an experimental model for the study of bladder contractility.
2007
Aims The objective of the study was to establish an experimental model for the extracorporeal perfusion of the pig detrusor. In order to validate this model we examined some biochemical parameters and determined the effect of carbachol on the contractility of perfused female pig bladders. Methods Twenty-six pig bladders were perfused with Krebs–Ringer bicarbonate-glucose buffer for a period up to 5 hr with the aim to preserve a viable organ, which would be responsive to contraction-inducing agents. The intravesical pressure of the bladder as well as the intraarterial pressure of the vesical arteries were recorded before and after administration of carbachol. Results The perfusate pH, lactat…
EFFECTS OF THIOBENCARB HERBICIDE TO AN ALGA (NANNOCHLORIS OCULATA) AND THE CLADOCERAN (DAPHNIA MAGNA)
2001
Chronic toxicity studies were conducted with an algae (Nannochloris oculata) and the cladoceran (Daphnia magna) to determine their relative sensitivities to the thiocarbamate herbicide thiobencarb (S-4-chlorobenzyl diethylthiocarbamate). Most of the algal populations were initially affected by exposure to the herbicide. Thiobencarb concentrations higher than 0.5 mg/L significantly reduced algal densities after 24-h exposure. The 24-h static EC50 in D. magna was 3.01 mg/L. The sublethal effects of 0.3, 0.37, 0.5, 0.75, and 1.5 mg/L of thiobencarb concentrations on the survival, reproduction, and growth of D. magna were monitored for 21 days. The parameters used to determined the effect of th…
Response and Recovery of Brain Acetylcholinesterase Activity in the European Eel,Anguilla anguilla,Exposed to Fenitrothion
1998
European eel (Anguilla anguilla) were exposed to sublethal fenitrothion concentrations in a continuous flow-through system for 4 days. Brain acetylcholinesterase (AChE) activity was evaluated after 2, 8, 12, 24, 32, 48, 56, 72, and 96 h pesticide exposure. Results indicated that AChE activity in eel brains decreased as the concentration of fenitrothion increased. The pesticide induced significant inhibitory effects on the AChE activity ofA. anguilla, ranging from >40% inhibition at a sublethal concentration of 0.02 ppm to >60% inhibition at a sublethal concentration of 0.04 ppm. Eel were exposed to both fenitrothion concentrations for 96 h and then allowed a period of recovery in pesticide-f…
Human Embryonic Stem Cell Derived Hepatocyte-Like Cells as a Tool for In Vitro Hazard Assessment of Chemical Carcinogenicity
2011
Hepatocyte-like cells derived from the differentiation of human embryonic stem cells (hES-Hep) have potential to provide a human relevant in vitro test system in which to evaluate the carcinogenic hazard of chemicals. In this study, we have investigated this potential using a panel of 15 chemicals classified as noncarcinogens, genotoxic carcinogens, and nongenotoxic carcinogens and measured whole-genome transcriptome responses with gene expression microarrays. We applied an ANOVA model that identified 592 genes highly discriminative for the panel of chemicals. Supervised classification with these genes achieved a cross-validation accuracy of > 95%. Moreover, the expression of the response g…
Induction of apoptosis and inhibition of cell growth in human hepatocellular carcinoma cells by COX-2 inhibitors
2005
The aim of the present study was to examine the effects of nonselective (indomethacin) and selective cyclooxygenase-2 (COX-2) inhibitors (NS-398, nimesulide, and CAY10404) on cell growth, cell cycle distribution, and apoptosis in three human hepatocellular carcinoma cell lines (HepG2, HuH-6, and HA22T/VGH) with different characteristics of differentiation and biological behavior. The four COX inhibitors showed a dose-dependent growth-inhibitory effect in all the cell lines. No substantial arrests in the progression of the cells through the cell cycle were observed after treatment of HuH-6 or HA22T/VGH for 48 h with the various inhibitors. On the other hand, there were significant increases …
Cytotoxic effects and degradation products of three mycotoxins: Alternariol, 3-acetyl-deoxynivalenol and 15-acetyl-deoxynivalenol in liver hepatocell…
2015
This work is focused in studying the cytotoxic effects on HepG2 cells of the mycotoxins alternariol (AOH), 3-acetyl-deoxynivalenol (3-ADON) and 15-acetyl-deoxynivalenol (15-ADON) by the MTT assay, as well as in the identification of the degradation products and/or metabolites originated after treatment by liquid chromatography tandem mass spectrometry (LC-MS/MS) equipment and extracted from culture media. HepG2 cells were treated at different concentrations over 24, 48 and 72 h. The IC50 values were from 65 to 96 μM, from 3.6 to 6.2 μM and from 5.2 to 8.1 μM for AOH, 3-ADON and 15-ADON, respectively. Among all three mycotoxins assayed, deoxynivalenol (DON) derivated presented the highest to…
Lasciare che l'altro sia. Uno sguardo fenomenologico-ermeneutico sull’autentico aver-cura.
2023
Starting from the authentic direction of care indicated by Heidegger, it is possible to trace a model of caring for the other that is non-intrusive and authentically paves the way for inclusiveness. It presents itself as a phenomenon whose essence is relationality: it is a phenomenal observation that tells us that being in the world is always a being among things and in relationship with others and care always takes place as a relationship that connects us in the world. The authentic direction of care is the one in which one knows how to conserve the other in its essence by guarding and cultivating it, the inauthentic interpretation is instead the one in which one relates to things and rela…
Exploring Finnish Guidance Counselors’ Conceptions of Career Management Skills
2020
This article reports the findings from a phenomenographic investigation into guidance counselors’ conceptions of career management skills (CMS). The results show that CMS was conceived as (a) information-based knowledge, (b) personal skills development, (c) interpersonal skills development, and (d) autonomous application of skills. The differences appeared along six dimensions of variation that included awareness of CMS, emphasis, promotion of CMS, teaching practice, assessment, and attitude. The findings give us a more profound understanding of critical aspects that may have an important role in the development of individual’s CMS.
‘I changed my life’s course’ : The impact of the Helsingin Sanomat Foundation fellowship on the career path of a journalist
2017
This study focuses on the fellowship program of Helsingin Sanomat Foundation, which has sent a hundred mid-career journalists from Finland to study abroad at prominent universities. Based on two surveys, one on the fellows (n = 45) and one on a control group (n = 52), the article discusses the impact the fellowship education has on the journalists and their careers. The surveys are complemented with in-depth interviews of 10 fellow journalists. The results are discussed in the context of protean career concept, and they indicate that the fellowship period has a notable positive impact on the career of an individual journalist, but a contradictory impact on the journalist’s home organizatio…
La première rencontre du corps malade en contexte de soins infirmiers : la relation de soin : une expérience ultime, du sensible au social
2016
Our study focuses on nursing care with a first approach based on human body and emotions through the teaching context in the sensitive hospital environment. The nursing student is a central point of our research as he lives a unique sensitive and interpersonal experience within his own body in a social setting imbued with symbolism. He perceives health care community through his five senses which inform and direct him, but also may destabilize him. We decided to base our study on the information and communication sciences thanks to a sensitive, sensorial and symbolic problematisation and through a multidisciplinary conceptualization based on different theoretical approaches, symbolic intera…
