Development of a simulation chamber for the evaluation of dermal absorption of volatile organic compounds

Abstract An exposition chamber has been designed and developed for the production of contaminated atmospheres with fixed and known concentration of volatile organic compounds. The proposed chamber has been evaluated for the determination of in-vitro diffusion parameters, such as flux and lag time for benzene, toluene, ethylbenzene, and xylenes (BTEX). Polydimethylsiloxane (PDMS) and Strat-M® membranes were evaluated, including the effect of membrane thickness, from 0.5 to 3.0 mm, on the diffusion parameters. A colorimetric composite device is proposed to determine the widely used biocide N-(3-aminopropyl)-N-dodecyl-1,3-propanediamine (ADP).This sensing device is based on a film of 1,2-Naphthoquinone-4-sulfonate (NQS) embedded into polydimethylsiloxane-tetraethylortosilicate-SiO2 nanoparticles composite (PDMS-TEOS-SiO2NPs). Semiquantitative analysis can be performed by visual inspection. Digitalized image or diffuse reflectance (DR) measurements can be carried out for quantitative analysis. Organ-on-chip (OOC) devices are miniaturized devices replacing animal models in drug discovery and toxicology studies. The majority of OOC devices are made from polydimethylsiloxane (PDMS), an elastomer widely used in microfluidic prototyping, but posing a number of challenges to experimentalists, including leaching of uncured oligomers and uncontrolled absorption of small compounds. Here we assess the suitability of polylactic acid (PLA) as a replacement material to PDMS for microfluidic cell culture and OOC applications. Polydimethylsiloxane (PDMS) is commonly used in medical devices because it is non-toxic and stable against oxidative stress. Relatively high blood platelet adhesion and the need for chemical crosslinking through curing, however, limit its utility. Polysiloxanes are useful materials because of their functionality, such as high thermal stability, electrical resistance, and hydrophobicity. In this regard, we studied palladium-catalyzed formation of iodo-and bromopolysiloxanes from two types of hydrosiloxanes, alpha,omega-dihydropoly(dimethylsiloxane) (1) and cyclotetra(hydromethylsiloxane) (2), and their reactions. Exposure assessments conducted using a personal sampler include the contribution of human activities to exposure that is neglected when using a stationary air sampler. Ordered networks of rat hippocampal neurons attached to silicon oxide surfaces.

The control of neuronal cell position and outgrowth is of fundamental interest in the development of applications ranging from cellular biosensors to tissue engineering. We have produced rectangular networks of functional rat hippocampal neurons on silicon oxide surfaces. Attachment and network formation of neurons was guided by a geometrical grid pattern of the adhesion peptide PA22-2 which matches in sequence a part of the A-chain of laminin. Abstract A series of commercial silicas and a laboratory made product were subjected to acidic/hydrothermal treatment with solutions of HCI, HNO3, H2SO4 and HF. The concentration of acid and the temperature and duration of treatment were varied. The specific surface area, content of metal impurities, types and concentration of surface hydroxyl and siloxane groups and crystallinity were determined for native and treated silicas.
