Germline correction of an epimutation related to Silver-Russell syndrome.
2015
Like genetic mutations, DNA methylation anomalies or epimutations can disrupt gene expression and lead to human diseases. However, unlike genetic mutations, epimutations can in theory be reverted through developmental epigenetic reprograming, which should limit their transmission across generations. Following the request for a parental project of a patient diagnosed with Silver-Russell syndrome (SRS), and the availability of both somatic and spermatozoa DNA from the proband and his father, we had the exceptional opportunity to evaluate the question of inheritance of an epimutation. We provide here for the first time evidence for efficient reversion of a constitutive epimutation in the sperm…
Silver nanoparticles stabilized by a polyaminocyclodextrin as catalysts for the reduction of nitroaromatic compounds
2015
Abstract Silver nanoparticles stabilized by means of poly -(6- N , N -dimethyl-propylenediamino)-(6-deoxy)-β-cyclodextrin were synthesized, characterized by different techniques (UV–vis spectroscopy, Dynamic Light Scattering, High Resolution Transmission Electron Microscopy, Fourier-transform IR Spectroscopy) and used as catalysts for the reduction of various nitrobenzene derivatives with sodium borohydride. The nanocomposites obtained appear to have an organized structure, with a metal core surrounded by a layer-structured coating shell. Kinetic data, rationalized in terms of a modified Langmuir–Hinshelwood model, evidenced a non-linear dependence of the reaction rate on the concentration …
Coffee Silverskin and Spent Coffee Suitable as Neuroprotectors against Cell Death by Beauvericin and α-Zearalenol: Evaluating Strategies of Treatment
2021
Coffee silverskin and spent coffee have been evaluated in a neuroblastoma cell line (SH-SY5Y cells) against beauvericin (BEA) and α-zearalenol (α-ZEL)-induced cytotoxicity with different strategies of treatment. First, the direct treatment of mycotoxins and coffee by-products extracts in SH-SY5Y cells was assayed. IC50 values for α-ZEL were 20.8 and 14.0 µM for 48 h and 72 h, respectively and, for BEA only at 72 h, it was 2.5 µM. Afterwards, the pre-treatment with spent coffee obtained by boiling water increased cell viability for α-ZEL at 24 h and 48 h from 10% to 16% and from 25% to 30%, respectively
Antiproliferative Properties of a Few Auranofin-Related Gold(I) and Silver(I) Complexes in Leukemia Cells and their Interferences with the Ubiquitin …
2020
A group of triethylphosphine gold(I) and silver(I) complexes, structurally related to auranofin, were prepared and investigated as potential anticancer drug candidates. The antiproliferative properties of these metal compounds were assessed against two leukemia cell lines, i.e., CCRF-CEM and its multidrug-resistant counterpart, CEM/ADR5000. Interestingly, potent cytotoxic effects were disclosed for both series of compounds against leukemia cells, with IC50 values generally falling in the low-micromolar range, the gold derivatives being on the whole more effective than the silver analogues. Some initial structure-function relationships were drawn. Subsequently, the ability of the study compo…
A proteomic approach to studying plant response to crenate broomrape (Orobanche crenata) in pea (Pisum sativum)
2004
Abstract Crenate broomrape ( Orobanche crenata ) is a parasitic plant that threatens legume production in Mediterranean areas. Pea ( Pisum sativum ) is severely affected, and only moderate levels of genetic resistance have so far been identified. In the present work we selected the most resistant accession available (Ps 624) and compared it with a susceptible (Messire) cultivar. Experiments were performed by using pot and Petri dish bioassays, showing little differences in the percentage of broomrape seed germination induced by both genotypes, but a significant hamper in the number of successfully installed tubercles and their developmental stage in the Ps 624 compared to Messire. The prote…
PROTEOMIC IDENTIFICATION OF NOVEL MARKERS IN BREAST AND COLON CANCER
Background: Discovery of new biomarker represent the greatest promise for the detection and management of cancer. Although progress in cancer biology has been rapid during the past few years, the complete understanding of molecular basis for cancer initiation, progression and efficacious treatments is still lacking. In this context, the application of proteomic strategies is now holding a focal position. The main reason is that proteins are the functional players that drive cancer phenotypes. Among cancers, breast and colon represent the most frequent forms. The evolution of these type of cancer are not easily predictable since there are several types that behave differently among patients.…
Inorganic/organic hybrid nanoparticles synthesized in a two-step radiation-driven process
2022
In this work, we have synthesized inorganic-organic hybrid nanoparticles via radiation synthesis of inorganic nanoparticles (Ag and CeO2) in aqueous dispersions containing radiation-synthesized poly(N-vinyl pyrrolidone) (PVP) nanogels (NG). The experiments show that there are strong interactions between the inorganic precursors (Ag+ and Ce3+) and the nanogel prior to irradiation. The two hybrid systems (Ag/NG and CeO2/NG) were characterized using X-ray diffraction (XRD) and transmission electron microscopy (TEM). XRD confirms the formation of crystalline Ag and CeO2. TEM reveals that the inorganic nanoparticles are evenly distributed in/on the nanogel. Both XRD and TEM show that size of the…
Electroreduction of aliphatic chlorides at silver cathodes in water
2010
The effect of water on the reduction of aliphatic chlorides at silver cathodes was investigated by cyclovoltammetric experiments in acetonitrile, water and their mixture and focused electrolyses in water solutions. The addition of water to acetonitrile gives rise to the formation of new peaks and/or the displacement of pre-existent peaks at dramatically less negative potentials. In a water medium, silver shows a very large electrocatalytic effect towards the reduction of chlorinated aliphatic compounds belonging to the classes of chloromethanes, chloroethanes and chloroethenes, dramatically higher than that observed in aprotic solvents such as dimethylformamide (DMF) or acetonitrile (ACN). …

2020
Plant extract of Pulicaria undulata (L.) was used as both reducing agent and stabilizing ligand for the rapid and green synthesis of gold (Au), silver (Ag), and gold&ndash
Effect of application time and concentration of silver diamine fluoride on the enamel remineralization
2021
Background Silver diamine fluoride has attracted attention because of its clinical success in arresting dental caries. Thus, the aim of this study was to evaluate the effect of different application times and concentrations of silver diamine fluoride (SDF) on deciduous tooth enamel remineralization. Material and Methods Blocks of deciduous tooth enamel were categorized into six groups of 11 each: 2 control groups: intact enamel, and demineralized enamel; 38% SDF and 30% SDF which were subdivided according to application times (1 and 3 min). The microhardness of samples was determined, and all groups except the intact enamel group were subjected to pH cycling to produce initial carious lesio…
