Compact Cell Imaging Device (CoCID) provides insights into the cellular origins of viral infections
2021
The overall CoCID concept is centred on providing virologists with a next-generation imaging device, which, through increased penetration and depth of focus, as well as through high natural contrast and sensitivity to organelle density (including virus-related organelles), will produce higher-fidelity ultrastructural images of whole intact cells. These insights will, in turn, help increase our understanding of the links between the structural reorganisation of cells and the mechanisms of viral entry, replication, assembly, and egress in cells. CoCID will provide this valuable imaging capability in the form of a compact lab-scale device that will greatly improve the accessibility of soft X-r…
G2/M checkpoint regulation and apoptosis facilitate the nuclear egress of parvoviral capsids
2022
The nuclear export factor CRM1-mediated pathway is known to be important for the nuclear egress of progeny parvovirus capsids in the host cells with virus-mediated cell cycle arrest at G2/M. However, it is still unclear whether this is the only pathway by which capsids exit the nucleus. Our studies show that the nuclear egress of DNA-containing full canine parvovirus. capsids was reduced but not fully inhibited when CRM1-mediated nuclear export was prevented by leptomycin B. This suggests that canine parvovirus capsids might use additional routes for nuclear escape. This hypothesis was further supported by our findings that nuclear envelope (NE) permeability was increased at the late stages…
Bio-imaging with the helium-ion microscope: A review
2021
Scanning helium-ion microscopy (HIM) is an imaging technique with sub-nanometre resolution and is a powerful tool to resolve some of the tiniest structures in biology. In many aspects, the HIM resembles a field-emission scanning electron microscope (FE-SEM), but the use of helium ions rather than electrons provides several advantages, including higher surface sensitivity, larger depth of field, and a straightforward charge-compensating electron flood gun, which enables imaging of non-conductive samples, rendering HIM a promising high-resolution imaging technique for biological samples. Starting with studies focused on medical research, the last decade has seen some particularly spectacular …

Consistency of Targeted Metatranscriptomics and Morphological Characterization of Phytoplankton Communities
2020
The composition of phytoplankton community is the basis for environmental monitoring and assessment of the ecological status of aquatic ecosystems. Community composition studies of phytoplankton have been based on time-consuming and expertise-demanding light microscopy analyses. Molecular methods have the potential to replace microscopy, but the high copy number variation of ribosomal genes and the lack of universal primers for simultaneous amplification of prokaryotic and eukaryotic genes complicate data interpretation. In this study, we used our previously developed directional primer-independent high-throughput sequencing (HTS) approach to analyze 16S and 18S rRNA community structures. C…
Protein purification of Agrobacterium tumefaciens phytochrome Agp1 and UV-vis spectroscopy analysis in comparison to DrBphP from Deinococcus Radiodur…
2015
Phytochromes are bimodal light switchable photosensors exist in plants, bacteria and fungi. Incorporating bilin as their chromophore, most bacteriophytochromes (BphPs) function as two-component histidine kinase affected by red/far-red light and regulate downstream physiological activities in the microorganism. The study aims on the purification of Agrobacterium phytochrome Agp1whose phosphorylation activity in its histidine kinase (HK) domain was recently found to be affected by the temperature. The purified proteins Agp1 were assembled with biliverdin IXα (BV) and monitored by UV-vis spectroscopy with BV incorporated DrBphP from Deinococcus radiodurans. Extinction coefficient spectra of “a…
Integrins as cellular receptors for fibril-forming and transmembrane collagens
2004
Regulation of raft-derived endocytic pathways : studies on echovirus 1 and baculovirus
2009

Collagen receptor integrins : evolution, ligand binding selectivity and the effect of activation
2007
Kollageeni on elimistössämme runsaimmin esiintyvä valkuaisaine, josta tunnetaan noin 30 alatyyppiä. Kollageenireseptori-integriinit sitoutuvat kolla…
