First spermatological data on the digenean genus Ityogonimus derived from the description of sperm characters of Ityogonimus ocreatus (Brachylaimidae…
2021
Abstract The present study describes the ultrastructural organization of the spermatozoa of the brachylaimid digenean Ityogonimus ocreatus (Ityogoniminae) by means of transmission electron microscopy (TEM). Live digeneans were collected from the digestive tract of an Iberian mole Talpa occidentalis (Eulipotyphla, Talpidae) captured accidentally during a vole pest control campaign in Priesca (Asturias, Spain). The TEM study reveals that the I. ocreatus sperm are filiform, tapered at both extremities, and have two 9+'1' trepaxonematan axonemes, external ornamentation of the plasma membrane associated with cortical microtubules, spine-like bodies, two bundles of parallel cortical microtubules …
Structural factors controlling ligand binding to myoglobin: a kinetic hole-burning study.
1998
Using temperature-derivative spectroscopy in the temperature range below 100 K, we have studied the dependence of the Soret band on the recombination barrier in sperm whale carbonmonoxy myoglobin (MbCO) after photodissociation at 12 K. The spectra were separated into contributions from the photodissociated species, Mb*CO, and CO-bound myoglobin. The line shapes of the Soret bands of both photolyzed and liganded myoglobin were analyzed with a model that takes into account the homogeneous bandwidth, coupling of the electronic transition to vibrational modes, and static conformational heterogeneity. The analysis yields correlations between the activation enthalpy for rebinding and the model p…
Comparison of different statistical approaches to evaluate morphometric sperm subpopulations in men.
2016
This study was designed to characterize morphometric sperm subpopulations in normozoospermic men by using different statistical methods and examining their suitability to classify correctly different sperm nuclear morphologies present in human ejaculates. Ejaculates from 21 normozoospermic men were collected for the study. After semen collection and analysis, samples were prepared for morphometric determination. At least 200 spermatozoa per sample were assessed for sperm morphometry by computer-assisted sperm morphometry analysis (CASA-Morph) using fluorescence. Clustering and discriminant procedures were performed to identify sperm subpopulations from the morphometric data obtained. Cluste…
DNA content in nine species of Nematocera with special reference to the sibling species of the Anopheles maculipennis group and the Culex pipiens gro…
1972
DNA values were determined for the nine species: Telmatoscopus meridionalis (family Psychodidae), Dixa obscura (family Dixidae), and Culiseta litorea, Culex pipiens, Aedes caspius, Anopheles labranchiae, Anopheles atroparvus, Anopheles stephensi, and Anopheles freeborni (family Culicidae). The DNA content indicated that the species could be divided into three categories: The Culex group with Culex pipiens, Culiseta litorea and Aedes caspius containing 1.02, 0.92 and 0.99 pg DNA in the haploid set, the Anopheles group including the four Anopheles species A. labranchiae, A. atroparvus, A. stephensi, A. freeborni and Telmatoscopus meridionalis with 0.23, 0.24, 0.24, 0.29 and 0.24 pg DNA and th…
Experimental Testicular Torsion: Effect on Endocrine and Exocrine Function and Contralateral Testicular Histology
1988
In order to investigate whether unilateral testicular torsion exerts a negative influence on the previously undisturbed contralateral side, exocrine and endocrine testicular function were evaluated before and two months after torsion. A rat model with 6 hours', 12 hours' or permanent extravaginal 540 degrees torsion of the right testis was used; a sham operated group of animals served as controls. Ejaculates were collected by electrostimulation; LH, FSH and testosterone serum levels were determined by radioimmunoassays. Eight weeks after torsion sperm output had decreased by half in the experimental groups, and LH levels increased significantly, whereas the other hormone levels, as well as …
Consequences on offspring of abnormal function in ageing gametes.
2000
The present review aims to analyse (i) the molecular, biochemical and cellular changes that accompany oocyte and sperm ageing in any of the internal or external environments where they can reside, and (ii) the consequences of the abnormal function in ageing gametes on pre- and post-implantation development and later life of offspring. This review also aims to propose and discuss cellular/molecular mechanisms framed within the 'free radical theory of ageing'. It appears that the ageing of gametes prior to fertilization may affect many molecular, biochemical and cellular pathways that may jeopardize not only pre- and post-implantation embryo/fetal development but also later life of offspring.…
New molecular markers for the evaluation of gamete quality.
2013
Purpose: Only 30 % of IVF cycles result in a pregnancy, so that multiple embryos need to be replaced, per treatment cycle, to increase pregnancy rates, resulting in a multiple gestation rate of 25 %. The use of new markers in the gamete selection, could reduce the number of the oocytes to be fertilized and embryos to be produced, but the tools to evidence the gamete competence remain unavailable and more studies are needed to identify bio-markers to select the best oocyte and sperm to produce embryos with higher implantation potentiality. Methods: To define oocyte competence, the apoptosis of the surrounding cumulus cells and the oxygen consumption rates for individual oocytes before fertil…
Comparison of three different staining methods for the assessment of epididymal red deer sperm morphometry by computerized analysis with ISAS®
2005
When collection of ejaculated sperm samples is not possible, as is the case with wild species, the epididymides of sacrificed wild males become the only possible source of spermatozoa. Mature cauda epididymal spermatozoa display characteristics similar to those of ejaculated sperm cells. The present work proposes a sperm staining technique suitable for the morphometric evaluation of red deer epididymal sperm using a new computerized system. Epididymides from wild animals were extracted no later than 2 h post mortem. After epididymal sectioning, sperm samples were collected, cooled to and equilibrated at 5 °C, and frozen in liquid nitrogen. Before staining, sperm samples were thawed for 20 s…
Virus-like particles, bacteria and microsporidia affect spindle-associated membranes in spermatocytes of Lepidoptera species.
1997
SummaryLarval testes of four Lepidoptera species were examined using electron microscopy. The testes of one species, the Mediterranean mealmothEphestia kuehniella(Pyralidae), were devoid of intracellular pathogens and serve as a control. In this species, metaphase spindles of primary spermatocytes showed a thick layer of perispindle membranes. The membranes were structurally very similar to the agranular endoplasmic reticulum. Membranes of this type occurred also at high frequency throughout the spindle matrix. The analysis of larval testes ofPieris brassicae(Pieridae) revealed virus-like particles within spermatocytes. In another species,Philudoria potatoria(Lasiocampidae), the spermatocyt…
Structural characterization and primary in vitro cell culture of locust male germline stem cells and their niche
2011
AbstractThe establishment of in vitro culture systems to expand stem cells and to elucidate the niche/stem cell interaction is among the most sought-after culture systems of our time. To further investigate niche/stem cell interactions, we evaluated in vitro cultures of isolated intact male germline–niche complexes (i.e., apical complexes), complexes with empty niche spaces, and completely empty niches (i.e., isolated apical cells) from the testes of Locusta migratoria and the interaction of these complexes with isolated germline stem cells, spermatogonia (of transit-amplifying stages), cyst progenitor cells, cyst progenitor cell-like cells, cyst cells, and follicle envelope cells. The stru…
