Microspore embryogenesis inCitrusand other fruit crops
2017
Conventional methods, involving several generations of selfing, are not applicable to produce homozygous lines in Citrus as well as in the other fruit crops, due to the high heterozygosity of the genomes, the long duration of the generation cycle, the large size, and, often, the self-incompatibility. For this reason, there is no different way to obtain homozygosity in this kind of plants than âgametic embryogenesisâ that allows the development of haploids (Hs, plants with gametophytic chromosome number) and doubled haploids (DHs, haploids that have undergone chromosome duplication) from heterozygous parents in a single step. Therefore, gametic embryogenesis is increasingly object of res…
Gametic embryogenesis through isolated microspore culture in mandarin (Citrus reticulata Blanco), Mandarino Tardivo Di Ciaculli: effect of meta-Topol…
2016
Haploid technology, allowing the single-step development of complete homozygous genotypes from heterozygous plants through gametic embryogenesis, has already an enormous impact on breeding programs of many important crops. Microspore embryogenesis can be carried out through in vitro culture of anther or isolated microspore. Experiments have been carried out via isolated microspore culture in Citrus reticulata (Blanco), Mandarino Tardivo di Ciaculli, investigating the influence of two culture media, of two plant growth regulator types and of two temperature treatments applied before the culture or just after the culture. After ten months of culture, for all the media tested, different struct…
STUDY ON GAMETIC EMBRYOGENESIS VIA IN VITRO ANTHER AND ISOLATED MICROSPORE CULTURE IN FRUIT CROPS
2014
Fruit breeding is mainly based on both conventional (hybridization, mutation and selection) or biotechnological methods (somatic hybridization, genetic transformation and haploid production). The genetic improvement through the conventional methods is limited by many factors such as fruit trees long juvenile period, high heterozygosity, large size and sexual incompatibility. Haploids and doubled haploids, obtained through gametic embryogenesis have a potential use in fruit crops genetic improvement. The change of the microspores fate from the normal gametopytic pathway towards the sporophytic induction is affected by numerous factors. Genotype, medium composition and stress were considered …

Resistance to space simulating conditions and sporicidal treatments of spores from bacilli of extreme environments origins: implication for Astrobiol…
Microorganisms able to tolerate environmental extremes, or extremophiles, are ideal candidates to extend our knowledge on the limitations for terrestrial life, including sporicidal treatments, and on their ability to survive under conditions mimicking space environments. The spore resistance of bacilli isolated from extreme environments, cold (Antarctic soils) and hot (shallow hydrothermal vents of Eolian Islands, Italy), was evaluated towards environmental stressors (wet- and dry-heat, low and high pH values), sporicidal treatments and stresses simulating space-environments (UV-C and X-rays irradiations, desiccations by low pressure, exposition to oxidizing agents and low pressure plasma),…
A study of the Candida albicans cell wall proteome
2008
Considering the importance of proteins in the structure and function of the cell wall of Candida albicans, we analyzed the cell wall subproteome of this important human pathogen by LC coupled to MS (LC-MS) using different protein extraction procedures. The analyzed samples included material extracted by hydrogen fluoride-pyridine (HF-pyridine), and whole SDS-extracted cell walls. The use of this latter innovative procedure gave similar data as compared to the analysis of HF-pyridine extracted proteins. A total of 21 cell wall proteins predicted to contain a signal peptide were identified, together with a high content of potentially glycosylated Ser/Thr residues, and the presence of a GPI mo…
Field distribution ofSorosphaera viticolain commercial vineyards in Germany
2006
In the year 2000, resting spores of a previously undescribed plasmodiophorid were found in roots of Vitis spp. This plasmodiophorid was identified as a member of the genus Sorosphaera Schroeter and described as Sorosphaera viticola Kirchmair, Neuhauser, Huber. To attain information on the field distribution of Sorosphaera viticola, a selective screening was conducted in two commercial vineyards in Germany. A study to determine a correlation of Sorosphaeraviticola infection to grapevine growth was also performed.
CRYPTIC SPECIES RELATED TO DALDINIA CONCENTRICA AND D. ESCHSCHOLZII, WITH NOTES ON D. BAKERI
2004
Daldinia macaronesica (from the Canary Islands and Madeira), D. palmensis (from the Canary Islands), D. martinii and D. raimundi (from Sicily), and D. vanderguchtiae (from Jersey, Channel Islands) spp. nov., are described, based on new combinations of teleomorphic and anamorphic characters. They all resemble the pantropical D. eschscholzii and/or the European D. concentrica with regard to teleomorphic characters and secondary metabolite profiles generated by analytical HPLC. The status of the newly described taxa was established by SEM of ascospores and microscopic studies of their anamorphs in comparison with various materials of the aforementioned known species. HPLC and SEM studies on th…
