Exome Sequencing Reveals VCP Mutations as a Cause of Familial ALS

Summary Using exome sequencing, we identified a p.R191Q amino acid change in the valosin-containing protein ( VCP ) gene in an Italian family with autosomal dominantly inherited amyotrophic lateral sclerosis (ALS). Mutations in VCP have previously been identified in families with Inclusion Body Myopathy, Paget disease, and Frontotemporal Dementia (IBMPFD). Screening of VCP in a cohort of 210 familial ALS cases and 78 autopsy-proven ALS cases identified four additional mutations including a p.R155H mutation in a pathologically proven case of ALS. Pain-evoked blink reflex

The electrically evoked blink reflex (BR) consists of an ipsilateral R1 component (R1) at 11 ms and two bilateral components R2 at 33 ms and R3 at 83 ms. It is still unclear whether the R2 is mediated by activation of tactile or nociceptive afferents. For testing the nociceptive hypothesis, nociceptors of the supraorbital nerve were selectively activated by infrared laser stimuli in 10 subjects. Only painful laser stimuli evoked a bilateral early polyphasic BR response (LR2) at 71 ms. Stimulation of infraorbital and mental nerve dermatomes was equally effective. Immune-mediated necrotizing myopathy is characterized by a specific Th1-M1 polarized immune profile.

Immune-mediated necrotizing myopathy (IMNM) is considered one of the idiopathic inflammatory myopathies, comprising dermatomyositis, polymyositis, and inclusion body myositis. The heterogeneous group of necrotizing myopathies shows a varying amount of necrotic muscle fibers, myophagocytosis, and a sparse inflammatory infiltrate. The underlying immune response in necrotizing myopathy has not yet been addressed in detail. The aim of this study was to compare the effects of isometric training performed with progressive versus rapid rate of contraction on the knee extensor neuromuscular properties over a 7-wk period.Sixteen healthy male subjects trained quadriceps femoris muscle in a leg extension machine three times a week during 7 wk. Changes in neuromuscular function after tasks involving control of EMG versus torque feedback of the same duration.

This study was designed to compare alterations in neuromuscular function after two tasks of similar duration involving the control of (1) torque level fixed at 40% maximal voluntary contraction (MVC) torque (torque task) and (2) EMG level when exerting 40% MVC torque on the knee extensor muscles. Ten healthy subjects volunteered to participate in two testing sessions separated by approximately 2 h. Neuromuscular adaptations and serum hormones in women during short-term intensive strength training

The effects were investigated in ten women of intensive heavy resistance strength training lasting for 3 weeks on electromyographic (EMG) activity, muscle cross-sectional area (CSA) and voluntary force production characteristics of leg extensor muscles. Blood samples for the determinations of serum hormones were taken from five of the subjects. Significant increases occurred in the higher force portions of the isometric force-time curve with an increase of 9.7 (SD 8.4)% (P less than 0.01) in maximal peak force. Energy use efficiency of livestock farms in a mountain area of Sicily

The research aimed to evaluate the performance (in terms of energy) of livestock farms located in a Mediterranean mountain area and characterized by different stocking rates. Farm data were collected from in-person interviews of farmers of 58 farms representative of the livestock farms of the Madonie and Nebrodi mountains area (Sicily, Italy), including several parameters related to farm characteristics, animal, crop and pasture management.
