Objectives To evaluate the main symptoms of knee osteoarthritis (OA) and tissue structure changes after a single dose bone marrow–derived mononuclear cell (BM MNC) intra articular injection. Detection of infection by hematopoietic stem and progenitor cells (HSPCs) is essential to replace myeloid cells consumed during the immune response. HSPCs express some functional pattern recognition receptors involved in the recognition of Candida albicans. In this context, our group has previously demonstrated that C. albicans yeasts induce proliferation and differentiation of HSPCs via TLR2 and Dectin-1. In the present PhD thesis, we used in vitro and ex vivo models of HSPC differentiation to investigate the functional consequences for mature myeloid cells of exposure of HSPCs to PAMPs or C. albicans yeasts. Milker’s nodules, also called paravaccinia, is a DNA virus transmitted from infected cows to humans. It results from contact with cattle, cattle byproducts, or fomites. Classified as an occupational disorder, those at risk of exposure include farmers, butchers, and agricultural tourists. The viral infection begins 5—15 days after inoculation as an erythematous-purple, round nodule with a clear depressed center, and a surrounding erythematous ring. While familiar to those in farming communities, the presence of the nodule may be concerning to others, particularly the immunosuppressed. Dotā darba uzdevumi: sniegt teorētisko bāzi par pašregulējošu sistēmu; meklēt pazīmes, kuras raksturo sociālo vidi caur šo sistēmu; noteikt kritērijus, pēc kuriem tiks veikta Jūrmalas, Daugavpils un Ventspils pilsētu pašvaldību gadījumu analīze, tā parādot, kā izvēlētie kritēriji izpaužas šo pašvaldību gadījumā. Normal tissue damage limits the efficacy of anticancer therapy. For anthracyclines, the clinically most relevant adverse effect is cardiotoxicity. The mechanisms involved are poorly understood and putative cardioprotectants are controversially discussed. Here, we show that the lipid-lowering drug lovastatin protects rat H9c2 cardiomyoblasts from doxorubicin in vitro. Protection by lovastatin is related to inhibition of the Ras-homologous GTPase Rac1. It rests on a reduced formation of DNA double-strand breaks, resulting from the inhibition of topoisomerase II by doxorubicin. Doxorubicin transport and reactive oxygen species are not involved. To investigate the potency of the topoisomerase II (topo II) poisons doxorubicin and etoposide to stimulate the DNA damage response (DDR), S139 phosphorylation of histone H2AX (γH2AX) was analyzed using rat cardiomyoblast cells (H9c2). Etoposide caused a dose-dependent increase in the γH2AX level as shown by Western blotting. By contrast, the doxorubicin response was bell-shaped with high doses failing to increase H2AX phosphorylation. Identical results were obtained by immunohistochemical analysis of γH2AX focus formation, comet assay-based DNA strand break analysis, and measuring the formation of the topo II-DNA cleavable complex.
