Cytosine methylation of tRNA-Asp by DNMT2 has a role in translation of proteins containing poly-Asp sequences

AbstractThe Dnmt2 RNA methyltransferase catalyses the methylation of C38 in the anticodon loop of tRNA-Asp, but the molecular role of this methylation is unknown. Here, we report that mouse aspartyl-tRNA synthetase shows a four to fivefold preference for C38-methylated tRNA-Asp. Consistently, a 30% reduced charging level of tRNA-Asp was observed in Dnmt2 knockout (KO) murine embryonic fibroblast cells. Gene expression analysis with fluorescent reporter proteins fused to an N-terminal poly-Asp sequence showed that protein synthesis of poly-Asp-tagged reporter proteins was reduced in Dnmt2 KO cells as well. This study compares the songs from the original, English-language version of the 2013 Disney film Frozen to those of the Dutch dubbed version, investigating how the thematic representation of love and fear differ between these versions. To support this inherently qualitative analysis, this study employs the triangle of aspects, an analytical model that identifies certain aspects and variables central to animated musical film dubbing, allowing a quantification of differences between dubbed versions. Background: Procollagen C-proteinase enhancer-1 (PCPE-1) is an extracellular glycoprotein that increases activity of certain zinc metalloproteinases involved in tissue development and repair. Results: PCPE-1 binds uniquely to the C-propeptide region of the procollagen molecule. Conclusion: PCPE-1 enhances proteolysis by binding solely to the procollagen C-propeptides. Significance: These data may lead to future applications in the development of antifibrotic therapies.
On the Influence of Grammars on Crossover in Grammatical Evolution

Standard grammatical evolution (GE) uses a one-point crossover (“ripple crossover”) that exchanges codons between two genotypes. The two resulting genotypes are then mapped to their respective phenotypes using a Backus-Naur form grammar. This article studies how different types of grammars affect the resulting individuals of a ripple crossover. We distinguish different grammars based on the expected number of non-terminals chosen when mapping genotype codons to phenotypes, \(B_{avg}\). The grammars only differ in \(B_{avg}\) but can express the same phenotypes. The paper proposes, starting from some certain or probable allusions (in part. a passage in Galen's Commentary on 'Epidemics' 2, only extant in Arabic translation), but also from some striking omissions ('Ancient medicine' is not mentioned in the history of hygiene that Galen traces out in the central chapters of Thrasybulus), to reconstruct the history of this significant ‘concealment’. This will also be done in the light of the numerous passages in the corpus in which Galen advocates, constantly linking it to Hippocrates, the ideal of medicine that is ‘physiologica’.

Using Attribute Grammars for Description of Inductive Inference Search Space

The problem of practically feasible inductive inference of functions or other objects that can be described by means of an attribute grammar is studied in this paper. In our approach based on attribute grammars various kinds of knowledge about the object to be found can be encoded, ranging from usual input/output examples to assumptions about unknown object's syntactic structure to some dynamic object's properties. We present theoretical results as well as describe the architecture of a practical inductive synthesis system based on theoretical findings.
