Two-photon (2P) excitation is a cornerstone approach widely employed in neuroscience microscopy for deep optical access and sub-micrometric-resolution light targeting into the brain. Two-photon excitation microscopy of tryptophan-containing proteins.

We have examined the feasibility of observing single protein molecules by means of their intrinsic tryptophan emission after two-photon excitation. A respiratory protein from spiders, the 24-meric hemocyanin, containing 148 tryptophans, was studied in its native state under almost in vivo conditions. In this specific case, the intensity of the tryptophan emission signals the oxygen load, allowing one to investigate molecular cooperativity. As a system with even higher tryptophan content, we also investigated latex spheres covered with the protein avidin, resulting in 340 tryptophans per sphere. Self-trapped exciton luminescence in crystalline α-quartz under two-photon laser excitation

Abstract The luminescence of pure crystalline α-quartz is studied under pulsed ArF laser excitation. The luminescence parameters obtained correspond well with those of self-trapped excitons (STEs) in α-quartz, indicating that the excitation process is two-photon. The efficiency of two-photon excitation is of the same order of magnitude as the one-photon excitation of sodium salicylate. The STE luminescence decay kinetics and their temperature dependence under photoexcitation were recorded with higher accuracy than previously. Line shape measurements of rubidium 5S-7S two-photon transition

We report the use of a digital lock to measure the line profile and center frequency of rubidium 5S-7S two-photon transitions with a cw laser referenced to an optical frequency comb. The narrow, two-photon transition, 5S-7S (760 nm), insensitive to first-order in a magnetic field, is a promising candidate for frequency reference.

Two-photon light-sheet microscopy for high-speed whole-brain functional imaging of zebrafish neuronal physiology and pathology

We present the development of a custom-made two-photon light-sheet microscope optimized for high-speed (5 Hz) volumetric imaging of zebrafish larval brain for the analysis of neuronal physiological and pathological activity. High-speed volumetric two-photon light-sheet microscopy is challenging to achieve, due to constrains on the signal-to-noise ratio. To maximize this parameter, we optimized our setup for high peak power of excitation light, while finely controlling its polarization, and we implemented remote scanning of the focal plane to record without disturbing the sample. The mechanical properties (hardness and elastic modulus) of organically modified silicate thin films can be finely tuned by varying the degree of alkylation and thus the fraction of six- and four-membered siloxane rings in the organosilica matrix. This opens the way to large tunability of parameters that are of crucial practical importance for films that are finding increasing application in numerous fields ranging from microelectronics to chemical sensing.
