Construction and validation of cDNA-based Mt6k-RIT macro- and microarrays to explore root endosymbioses in the model legume Medicago truncatula

2004

To construct macro- and microarray tools suitable for expression profiling in root endosymbioses of the model legume Medicago truncatula, we PCR-amplified a total of 6048 cDNA probes representing genes expressed in uninfected roots, mycorrhizal roots and young root nodules [Nucleic Acids Res. 30 (2002) 5579]. Including additional probes for either tissue-specific or constitutively expressed control genes, 5651 successfully amplified gene-specific probes were used to grid macro- and to spot microarrays designated Mt6k-RIT (M. truncatula 6k root interaction transcriptome). Subsequent to a technical validation of microarray printing, we performed two pilot expression profiling experiments usin…

Developmental analysis of merosity and sexual morphs in Rubiaceae: A case study in Rubia and Cruciata

2016

The Rubiaceae is characterized by considerable diversity in floral features. To shed light on the variation of floral developmental patterns in Rubiaceae, a comparative study was carried out on pentamerous flowers of Rubia tinctorum L. and tetramerous flowers of Cruciata (Cruciata laevipes Opiz.; Cruciata glabra (L.) Ehrend.), two herbaceous genera of the tribe Rubieae. We aim to study the mechanism responsible for meristic variation and to discuss the expression of andromonoecy in C. laevipes. Developmental peculiarities include the time overlap in the initiation of petal and stamen whorls in Cruciata, the lack of calyx, and a very weak sympetaly in all species examined. Our results show t…

An isoleucine-leucine substitution in chloroplastic acetyl-CoA carboxylase from green foxtail (Setaria viridis L. Beauv.) is responsible for resistan…

2002

The cDNAs encoding chloroplastic acetyl-CoA carboxylase (ACCase, EC 6.4.1.2) from three lines of Setaria viridis (L. Beauv.) resistant or sensitive to sethoxydim, and from one sethoxydim-sensitive line of Setaria italica (L. Beauv.) were cloned and sequenced. Sequence comparison revealed that a single isoleucine-leucine substitution discriminated ACCases from sensitive and resistant lines. Using near-isogenic lines of S. italica derived from interspecific hybridisation, we demonstrated that the transfer of the S. viridis mutant ACCase allele into a sethoxydim-sensitive S. italica line conferred resistance to this herbicide. We confirmed this result using allele-specific polymerase chain rea…

Effects of starvation on survival, cannibalism, body mass, and intestinal protozoan profile in the subterranean termite Reticulitermes lucifugus

2019

Scarcity or inadequate nutrition can affect biological and behavioural aspects of subterranean termites and their intestinal protozoan profile. The aim of this study was to investigate changes in survival, cannibalism, body mass, and protist community structure of Reticulitermes lucifugus Rossi subspecies “Sicily” following starvation to provide basic knowledge for the development of more specific studies on a possible survival strategy under stressful conditions. In nature, this termite consumes many food sources and its feeding activity is continuous during the year. In a 35-day laboratory experiment, groups of 50 termites (worker/soldier ratio 49:1) were subjected to two diets, starvatio…

Application of anti-transpirant to control sugar accumulation in grape berries and alcohol degree in wines obtained from thinned and unthinned vines …

2020

In the last few decades, the accumulation of sugar in grape berries and hence the alcohol degree of wines has been affected by increasing global temperatures. In order to limit plant photosynthetic activity, it is possible to apply anti-transpirant on field, reducing sugar accumulation in berries. In this contest, our aim was to evaluate the efficiency of the application of a di-1-p-menthene-based natural anti-transpirant (Vapor Gard&reg

Tettigoniidae (Orthoptera) ovipositing in old galls of Dryocosmus kuriphilus (Hymenoptera: Cynipidae)

2016

Authors report some biological notes on two species of Orthoptera Tettigoniidae emerged from old spongy-woody galls of Dryocosmus kuriphilus Yasumatsu, 1951 collected in April 2015 from some areas of Sicily (Italy): Leptophyes sicula Kleukers, Odé et Fontana, 2010 (Phaneropterinae) and Cyrtaspis scutata (Charpentier, 1825) (Meconematinae). Between the end of April and the first days of May 30 neanids emerged from the galls, were reared and their cycle followed. While L. sicula laid eggs in groups, C. scutata laid single eggs inside the galls; both species have shown that in a few years they adapted in exploiting this new shelter for egg laying. No interaction with the gall inducing insect w…

Festuca valesiacaSchleich. ex Gaudin newly discovered in the Central Apennines (Italy): a further example of steppe relict in the Abruzzo “dry valley…

2019

Festuca valesiaca Schleich. ex Gaudin subsp. valesiaca is acknowledged as one of the most characteristic grasses of the continental grasslands and steppes of Eurasia. In Italy, the species was known almost only from the continental valleys of the Alpine chain (with the exception of two small populations at the northernmost edge of the Apennines). Here, we report on the first confirmed finding of F. valesiaca subsp. valesiaca in the surroundings of the Fucino Basin, Central Apennines. This finding expands by c. 400 km southwards the species’ range in Italy. The identity of the species was first ascertained by means of leaf cross-sections and stomata measurements, and then confirmed by chromo…

Osmotrophic glucose and leucine assimilation and its impact on EPA and DHA content in algae

2020

The uptake of dissolved organic compounds, that is, osmotrophy, has been shown to be an efficient nutritional strategy for algae. However, this mode of nutrition may affect the biochemical composition, for example, the fatty acid (FA) contents, of algal cells. This study focused on the osmotrophic assimilation of glucose and leucine by selected seven algal strains belonging to chlorophytes, chrysophytes, cryptophytes, dinoflagellates and euglenoids. Our laboratory experiments with stable isotope labeling showed that osmotrophy occurred in four of the selected seven strains. However, only three of these produced long chain omega-3 FAs eicosapentaenoic acid (EPA; 20:5ω3) and docosahexaenoic a…

A validity and reliability study of Conditional Entropy Measures of Pulse Rate Variability

2019

In this work, we present the feasibility to use a simpler methodological approach for the assessment of the short-term complexity of Heart Rate Variability (HRV). Specifically, we propose to exploit Pulse Rate Variability (PRV) recorded through photoplethysmography in place of HRV measured from the ECG, and to compute complexity via a linear Gaussian approximation in place of the standard model-free methods (e.g., nearest neighbor entropy estimates) usually applied to HRV. Linear PRV-based and model-free HRV-based complexity measures were compared via statistical tests, correlation analysis and Bland-Altman plots, demonstrating an overall good agreement. These results support the applicabil…
