Negative Regulation of β Enolase Gene Transcription in Embryonic Muscle Is Dependent upon a Zinc Finger Factor That Binds to the G-rich Box within th…

1998

We have previously identified a muscle-specific enhancer within the first intron of the human beta enolase gene. Present in this enhancer are an A/T-rich box that binds MEF-2 protein(s) and a G-rich box (AGTGGGGGAGGGGGCTGCG) that interacts with ubiquitously expressed factors. Both elements are required for tissue-specific expression of the gene in skeletal muscle cells. Here, we report the identification and characterization of a Kruppel-like zinc finger protein, termed beta enolase repressor factor 1, that binds in a sequence-specific manner to the G-rich box and functions as a repressor of the beta enolase gene transcription in transient transfection assays. Using fusion polypeptides of b…

Cancer relapse under chemotherapy: why TLR2/4 receptor agonists can help.

2007

Liver or lung metastases usually relapse under chemotherapy. Such life-threatening condition urgently needs new, systemic anticancer compounds, with original and efficient mechanisms of action. In B16 melanoma mice treated with cyclophosphamide, D'Agostini et al. [D'Agostini, C., Pica, F., Febbraro, G., Grelli, S., Chiavaroli, C., Garaci, E., 2005. Antitumour effect of OM-174 and Cyclophosphamide on murine B16 melanoma in different experimental conditions. Int. Immunopharmacol. 5, 1205-1212.] recently found that OM-174, a chemically defined Toll-like receptor(TLR)2/4 agonist, reduces tumor progression and prolongs survival. Here we review 149 articles concerning molecular mechanisms of TLR2…

Relationships between oxygen concentration and patterns of energy metabolism in the rotifer Brachionus plicatilis

1992

Abstract 1. 1. Activities of 10 enzymes and accumulation of two metabolites under experimental hypoxia were investigated in the rotifer Brachionus plicatilis . 2. 2. Pyruvate kinase (PK), lactate dehydrogenase (LDH), malic enzyme (ME), phosphoenolpyruvate carboxykinase (PEPCK), malate dehydrogenase (MDH) and fumarase (FUM) activities showed great increase at low oxygen levels. Alcohol dehydrogenase (ADH) activity was not detected. 3. 3. Alanine concentration showed an increase with low oxygen concentrations, while alanine aminotransferase (GTP) showed a slight activity increase with oxygen levels. The high alanine concentration was interpreted as enhanced proteolysis. 4. 4. Our results show…

An alternative allergen:adjuvant formulation potentiates the immunogenicity and reduces allergenicity of a novel subcutaneous immunotherapy product f…

2012

Background Subcutaneous specific immunotherapy (SCIT) has proven sustained clinical efficacy against allergy. The recommended regimen for SCIT is a gradual updosing over a period of weeks. Commonly, in commercial products for SCIT, the specific allergen is formulated with an adjuvant, most often in the form of aluminium hydroxide (AlOH). It has been shown that allergen-specific IgG antibodies are induced as a result of successful SIT. Objective To investigate the possibility of optimizing the formulation of AlOH-based grass-pollen allergy vaccines for SCIT in a way that allows for shorter updosing regimens while maintaining the immunogenicity of the vaccine. Methods Mice were immunized with…

Cellular stress induces cap-independent alpha-enolase/MBP-1 translation.

2015

AbstractMyc promoter-binding protein-1 (MBP-1) is a shorter protein variant of the glycolytic enzyme alpha-enolase. Although several lines of evidence indicate that MBP-1 acts as a tumor suppressor, the cellular mechanisms and signaling pathways underlying MBP-1 expression still remain largely elusive. To dissect these pathways, we used the SkBr3 breast cancer cell line and non-tumorigenic HEK293T cells ectopically overexpressing alpha-enolase/MBP-1. Here, we demonstrate that induced cell stresses promote MBP-1 expression through the AKT/PERK/eIF2α signaling axis. Our results contribute to shedding light on the molecular mechanisms underlying MBP-1 expression in non-tumorigenic and cancer c…

A rietveld study of the cation substitution between uvarovite and yttrium-aluminum synthetic garnets, obtained by sol-gel method

1994

A series of synthetic garnets solid solutions with compositions between Y 3 Al 2 Al 3 O 12 (Y-Al garnet) (YAG) and Ca 3 Cr 2 Si 3 O 12 (uvarovite) was synthesized using the sol-gel method. The expected general formula is (Ca x Y 1-x ) 3 (Cr x Al 1-x ) 2 (Si x Al 1-x ) 3 O 12 where x=0 to x=1. The cation distribution in dodecahedral, octahedral and tetrahedral sites and bond distances in these synthetic garnets were determined using the Rietveld method. It shows the incomplete substitution in small sites. The smaller site the smaller substitution there is
