Synthetic antigen-encoding mRNA is increasingly exploited as a tool for delivery of genetic information of complete antigens into professional antigen presenting dendritic cells for HLA haplotype-independent antigen-specific vaccination against cancer. Two strategies for mRNA-based antitumor vaccination have emerged into the clinical setting. One is transfection of autologous dendritic cells with synthetic mRNA for adoptive transfer into the patient. The other is direct injection of naked synthetic mRNA. International audience; TBVAC2020 is a research project supported by the Horizon 2020 program of the European Commission (EC). It aims at the discovery and development of novel tuberculosis (TB) vaccines from preclinical research projects to early clinical assessment. The project builds on previous collaborations from 1998 onwards funded through the EC framework programs FP5, FP6, and FP7. It has succeeded in attracting new partners from outstanding laboratories from all over the world, now totaling 40 institutions. Synthetic mRNA has emerged as a powerful tool for the transfer of genetic information, and it is being explored for a variety of therapeutic applications. Many of these applications require prolonged intracellular persistence of mRNA to improve bioavailability of the encoded protein. mRNA molecules are intrinsically unstable and their intracellular kinetics depend on the UTRs embracing the coding sequence, in particular the 3′ UTR elements. We describe here a novel and generally applicable cell-based selection process for the identification of 3′ UTRs that augment the expression of proteins encoded by synthetic mRNA. Method Is a retrospective observational study of vaccination with tetanus–diphtheria (Td) in adults of mutual injuries of work of Spain (FREMAP) between January 2010 and December 2013. The aim of this essay is to reflect on the governance of unvaccinated Italian children aged between zero and six years, due to the new regulatory system on mandatory vaccination in Italy (Law Decree no. 73/2017, converted into Law no. 119/2017). In order to strengthen and protect the right to health, the Italian government first, and then the lawmakers, set forth a specific prohibition preventing unvaccinated children from attending preschool, whether public or private. The first global report on health and ageing published by the World Health Organization (WHO) in 2015 introduced a new and innovative concept, namely, the idea that healthy ageing encompasses more than just the absence of disease. The report considered healthy ageing to be a more holistic process of developing and maintaining the functional ability that enables well-being in older age. There are two major components to this concept, namely, personal and environmental determinants of ageing. Abstract Vaccination is one of the most effective medical interventions ever introduced, preventing millions of cases of infections worldwide every year. However, vaccines are commonly believed to be less effective in providing protection in older adults, due to the perceived decline seen in immunity in this population (“immunosenescence”). Accordingly, this chapter considers how to identify hallmarks of immunosenescence and approach their resolution strategically. It was reviewed potential options for therapeutic intervention to restore appropriate responses to vaccines in older adults.
