Voltage gradient electrophoresis of nucleic acids on agarose gels.

A very simple method is described which allows the separation of DNA molecules in a wide molecular weight range (from 0.6 to about 30 kb) in the same electrophoresis agarose gel. This is based on the achievement of a voltage gradient through a simple device consisting of a Plexiglas plate placed slantwise with respect to the gel surface plane, submerged in the electrophoretic running buffer. Further applications of our system are also described.

Quantitative RT-PCR measurement of human cytochrome P-450s: application to drug induction studies.

A quantitative RT-PCR assay has been developed that is able to measure the mRNA content of the major human CYPs (1A1, 1A2, 2A6, 2B6, 2C9, 2C19, 2D6, 2E1, 3A4, and 3A5). The technique is highly specific, reproducible, rapid, and sensitive enough to quantitate low and high abundant mRNAs. The PCR primers were selected to specifically match each CYP mRNA, to have a very close annealing temperature, and to render PCR products of similar sizes. The PCR conditions were designed to allow the simultaneous measurement of the various human liver CYPs in a single run. Semi-automatic quantitative RT-PCR to measure CYP induction by drugs in human hepatocytes

An assay has been developed for the quantitative measurement of CYP mRNA content of the major human isoforms (1A1, 1A2, 2A6, 2B6, 2C9, 2C19, 2D6, 2E1, 3A4 and 3A5) in human hepatocytes. The method is based on the conversion of mRNAs into their corresponding cDNAs, followed by PCR amplification using appropriate primers. Making use of appropriate internal and external standards it is possible to estimate changes in CYP mRNA content of hepatocytes. The technique has been standardised to run semi-automatically. The Role of Assumptions in Ohlson Model Performance: Lessons for Improving Equity-Value Modeling

In this paper, we test whether the short-run econometric conditions for the basic assumptions of the Ohlson valuation model hold, and then we relate these results with the fulfillment of the short-run econometric conditions for this model to be effective. Better future modeling motivated us to analyze to what extent the assumptions involved in this seminal model are not good enough approximations to solve the firm valuation problem, causing poor model performance. This paper develops a social insurance accounting model for a notional defined contribution (NDC) scheme combining retirement and long-term care (LTC) contingencies. The procedure relies on standard double-entry bookkeeping and enables us to compile a “Swedish” type actuarial balance sheet (ABS) following a framework equivalent to an open group approach. LKS-92 Coordinates Transformation to ITRF2000

LKS-92 is Latvian geodetic coordinate system used as an EUREF89 realization in Latvia. LKS-92 is official coordinate system for procuring of essential geospatial data. For aeronautical and other purposes ITRF2000 is used in publication of geospatial data. Research is done to obtain transformation parameters from LKS-92 realization epoch 1992.75 through LatPos to ITRF2000 epoch 2000.00. Valuation of two different coordinate adjustment strategies and accuracy of parameters is done. Results of research could be used for transformation from LKS-92 to ITRF2000 for all kind of geospatial data.
