Even though it is known for more than one decade that antigen-encoding RNA can deliver antigenic information to induce antigen-specific immunity against cancer, the nature and mechanism of RNA uptake have remained enigmatic. In this study, we investigated the pharmacokinetics of naked RNA administered into the lymph node. We observed that RNA is rapidly and selectively uptaken by lymph node dendritic cells (DCs). We investigate the dynamics of a wild-type viral strain which generates mutant strains differing in phenotypic properties for infectivity, virulence, and mutation rates. We study, by means of a mathematical model and bifurcation analysis, conditions under which the wild-type and mutant viruses, which compete for the same host cells, can coexist. The coexistence conditions are formulated in terms of the basic reproductive numbers of the strains, a maximum value of the mutation rate, and the virulence of the pathogens. In a recent issue of Molecular Ecology the role of intragenomic parasites in maintaining sexual reproduction was both experimentally evaluated by Kraaijeveld et al. and discussed by Crespi and Schwander. The prevalence of sex is difficult to explain, due to its costs when compared with asexual reproduction. Yet, as reviewed by Crespi and Schwander, sex can be favorable in the presence of proliferating transposons. Transposons are similar to mutations, in that their integration to non-neutral loci is likely to have deleterious effects, and sexual recombination provides a potential mechanism to confine their accumulation.

This study evaluates the extent to which genetic differences among host individuals from the same species conditions the evolution of a plant RNA virus. We performed a three-fold replicated evolution experiment in which Tobacco etch potyvirus isolate At17b (TEV-At17b), adapted to Arabidopsis thaliana ecotype Ler-0, was serially passaged in five genetically heterogeneous ecotypes of A. thaliana. Many pathogens, including viruses, bacteria, as well as bacterial toxins, enter their target cells by endocytosis leading to accumulation of pathogenic and cellular proteins in endosomes. Here, we present detailed experimental instructions on isolation of endosomes after virus infection and their subsequent biomolecular characterization.
