In vitro reconstruction of an alveolar barrier for modeling normal lung functions and pathological events serve as reproducible, high-throughput pharmaceutical platforms for drug discovery, diagnosis, and regenerative medicine. Reactive Self-Assembly and Specific Cellular Delivery of NCO-sP(EO-stat-PO)-Derived Nanogels
This study presents the reactive self-assembly of isocyanate functional and amphiphilic six-arm, star-shaped polyether prepolymers in water into nanogels. Intrinsic molecular amphiphilicity, mainly driven by the isophorone moiety at the distal endings of the star-shaped molecules, allows for the preparation of spherical particles with an adjustable size of 100-200 nm by self-assembly and subsequent covalent cross-linking without the need for organic solvents or surfactants. Covalent attachment of a fluorescence dye and either the cell-penetrating TAT peptide or a random control peptide sequence shows that only TAT-labeled nanogels are internalized by HeLa cells. Special Families of Curves, of Abelian Varieties, and of Certain Minimal Manifolds over Curves
This survey article discusses some results on the structure of families f:V-->U of n-dimensional manifolds over quasi-projective curves U, with semistable reduction over a compactification Y of U. We improve the Arakelov inequality for the direct images of powers of the dualizing sheaf. For families of Abelian varieties we recall the characterization of Shimura curves by Arakelov equalities. For families of curves we recall the characterization of Teichmueller curves in terms of the existence of certain sub variation of Hodge structures. Main Chain Conformation and Anomalous Elution Behavior of Cylindrical Brushes As Revealed by GPC/MALLS, Light Scattering, and SFM
High molar mass polymacromonomers based on methacryloyl end-functionalized oligo methacrylates (Mn = 2410 g/mol) adopt the conformation of wormlike cylindrical brushes. Comparison of the absolute molar mass, Mw, determined by static light scattering and the contour length, Lw, of the molecules measured by SFM in the dry state revealed the length per vinylic main chain monomer of the cylindrical structure to be less than 0.1 nm, thus being much shorter than the maximum value of 0.25 nm. In solution this shrinkage could be quantified to 0.071 nm per monomer by Holtzer analysis of the scattering curves which in addition yielded the Kuhn statistical segment length lk = 120 nm. Targeted delivery of drugs across endothelial barriers remains a formidable challenge, especially in the case of the brain, where the blood-brain barrier severely limits entry of drugs into the central nervous system. Nanoparticle-mediated transport of peptide/protein-based drugs across endothelial barriers shows great potential as a therapeutic strategy in a wide variety of diseases. Functionalizing nanoparticles with peptides allows for more efficient targeting to specific organs.
