Intravenous BQ-123 and phosphoramidon reduce ventricular ectopic beats and myocardial infarct size in dogs submitted to coronary occlusion and reperf…

2001

Abstract The aim of this work was to investigate the influence of endothelin on myocardial ischemia and reperfusion in anaesthetized dogs. Animals were submitted to left thoracotomy and 120 min of left anterior descending coronary occlusion, followed by 180 min of reperfusion. Arterial blood pressure and electrocardiogram (ECG) were recorded in order to analyze heart rate (HR)–pressure product and production of ectopic beats. Infarcted areas were identified by a macroscopic staining method and infarct size was expressed as percentage of risk zone. To inhibit the effects of endothelin in a group of animals, we administered intravenously an endothelin synthesis inhibitor (phosphoramidon) and …

Synthesis and characterization of NdNiO3 prepared by low temperature methods

1992

Abstract NdNiO3 has been prepared with an orthorhombic perovskite structure by a low temperature and low oxygen pressure method starting from a nickel(III) oxide precursor, β-NiO(OH). The compound LaNiO3 has also been obtained by this method. This chemical-precursor-based synthesis overcomes problems related to the use of high oxygen pressures in the synthesis.

Contraction of human airways by oxidative stress

1999

We examined the in vitro effects of tert-butylhydroperoxide (tBu-OOH) in human bronchial muscle. tert-Butylhydroperoxide produced concentration-dependent contractions of bronchial rings (maximum effect was 56.5 +/- 9.6% of contraction by 1 mM acetylcholine; effective concentration 50% was approximately 100 microM). tert-Butylhydroperoxide (0.5 mM)-induced contraction was enhanced by epithelial removal but abolished by indomethacin (cyclooxygenase inhibitor) and zileuton (lipoxygenase inhibitor). tert-Butylhydroperoxide produced a transient rise in intracellular calcium in human cultured airway smooth muscle cells (HCASMC). The bronchial reactivity to acetylcholine and histamine was not alte…

Precursor-based synthetic pathways to nanometer NdNiO3−x particles

1993

Abstract The oxygen deficient neodymium nickel perovskite, NdNiO3−x, has been obtained by different low temperature-low oxygen pressure synthetic procedures. Besides a solid-matrix based route, both ordered and disordered chemical-precursors have been used as starting products. X-ray powder diffraction shows that the structure of this defective perovskite is orthorhombic. The study of the grain morphology of the products as resulting from the different synthetic procedures indicates that the main parameter determining the average particle size, which is in the nanometer range, is the maximum temperature achieved in the synthesis. Resistivity and DSC measurements show the existence of a meta…

Role of nuclear glutathione as a key regulator of cell proliferation.

2009

Glutathione (GSH) is essential for survival of eukaryotic but not in prokaryotic cells. Its functions in nucleated cells are far from being known. In fact GSH plays an important role in cell proliferation. The purpose of the present review is to summarize the relationship between glutathione and the important events that take place in the nucleus during the cell cycle. Most GSH co-localizes with nuclear DNA when cells are proliferating. However, when cells were confluent no differences between nucleus and cytoplasm could be seen. A number of relevant nuclear proteins are strictly dependent on nuclear redox status. For instance, we found that telomerase is regulated by shifts in glutathione …

Identification and expression of the 11β‐steroid hydroxylase from Cochliobolus lunatus in Corynebacterium glutamicum

2019

Hydroxylation of steroids has acquired special relevance for the pharmaceutical industries. Particularly, the 11β-hydroxylation of steroids is a reaction of biotechnological importance currently carried out at industrial scale by the fungus Cochliobolus lunatus. In this work, we have identified the genes encoding the cytochrome CYP103168 and the reductase CPR64795 of C. lunatus responsible for the 11β-hydroxylase activity in this fungus, which is the key step for the preparative synthesis of cortisol in industry. A recombinant Corynebacterium glutamicum strain harbouring a plasmid expressing both genes forming a synthetic bacterial operon was able to 11β-hydroxylate several steroids as subs…
