The effects of i.v. administered hypertonic solutions (6-fold isotonic solutions of NaCl, sodium sulfate, glucose and mannitol) theophylline (25 mg/kg) and vasopressin (50 mE/kg) on the blood pressure, cortical kidney vessel blood content and tubulus dilatation were studied in large groups of mice. The blood pressure and blood content effects after i.p. injection of 10% NaCl solution and the blood content during water diuresis were also investigated. The kidney vessel blood content was determined through benzidine stained erythrocytes in kidney slices. An increase in blood content as compared to corresponding controls was consistently noted after i.v. injection of 6-fold isotonic solutions and vasopressin and after i.p. injection of 10% NaCl. Blood distribution in the kidney cortex of animals at the peak of water diuresis and after theophylline did not differ from that of controls. Mannitol and sodium sulfate produced a marked dilatation of the tubules. The effect of theophylline, vasopressin and hypertonic NaCl and glucose solutions did not differ from the controls. Pronounced blood pressure effects were produced by sodium sulfate and vasopressin; hypertonic NaCl and glucose solutions were less effective, mannitol had no effect. Theophylline (25 mg/kg) produced an average blood pressure drop of 10±1,3 mm. Hg. Intraperitoneal injection of 2 ml. 10% NaCl solution caused an immediate rise in blood pressure (30–40 mm. Hg) followed by a constant decrease in the following 5–10 min. until the death of the animal. The results are discussed.
