The Role of HSP70 in the Diagnosis of HCC

ABSTRACT: Hepatocellular carcinoma (HCC) is a common tumor with a fatal clinical outcome if not diagnosed at an early stage. To date there is still a lack of reliable tumor biomarkers to detect asymptomatic precursor lesions in early HCC. Conventional pathological analysis also often fails to achieve sufficient sensitivity and specificity to diagnose early HCC. Genetic, proteomic, immunohistochemical, and liquid biopsy analysis indicate a role for members of the heat-shock protein 70 (HSP70) family as potential tumor-specific markers for HCC. Bacillus thuringiensis treatment modulate the HSP70 expression in larva and adult brain of Rhynchophorus ferrugineus

To study the pathogen-host relationship, we used the model of the entomopathogenic bacterium Bacillus thuringiensis (Bt) and Rhynchophorus ferrugineus, a quarantine pest that attacks palm trees. In particular, we focused on the Bt stress-induced infections. We studied the effect of Bt on larval and adult growth, and on the expression of the heat shock proteins (HSPs), rapidly synthesized in the cell after exposure to stress including pathogens. Bt has negative effects on larval and adult growth, on total hemocytes counts and on the hemocyte type. Localization of constitutive hsp70 in eggs and early embryos of sea urchin Paracentrotus lividus is shown by means of in situ immunostaining. An accumulation of this protein is shown in the mitotic structures (asters, spindles and centrosomes). Microinjection of anti-hsp70 antibodies into eggs causes impairment of formation of mitotic structures and of cell division. This impairment goes from a complete mitotic block, to irregular mitotic apparatus formation with irregular cleavage, depending upon the antibody concentration. Osteoblastic-like cells (SAOS-2) were seeded on three differently treated surfaces of Ti- 6Al-4V (T1: not treated; T2: chemically treated; T3: electrochemically treated). After 6h, the cells grown on T1 and T2 surfaces showed a typical poligonal morphology, they were strongly adherent to the surface and after 24h-48h they spread and formed a confluent monolayer. On the contrary, cells seeded on T3 showed heterogeneous morphology, decreased adhesion, loss of thin cellular expansions, and showed cellular shrinkage and apoptotic blebbing; only after 48h they were able to attach and form a monolayer after 4 days of seeding. Titanium is one of the most widely used biomaterials in orthopaedic, dental and trauma surgery. Superficial adhesion and cell proliferation represent the first steps of cell-biomaterial interactions. The efficacy of this early phase influences the subsequent ability to differentiate, and hence the knowledge of these cell activities is important for studying and improving the biocompatibility of biomaterials. The aim of this study was to test the adaptive ability of osteoblastic cells grown on titanium surfaces, including morphologic, proliferative and adaptative aspects.

Complete sequence of HSP70 cDNA from the mussel, Mytilus galloprovincialis was established before quantifying its expression following moderate heat shock or injection of heat-killed bacteria. HSP70 cDNA is comprised of 2378 bp including one ORF of 654 aa, with a predicted 70 bp 5'-UTR and a 343 bp 3'-UTR (GenBank, 18 Jan 05, AY861684). Alignment identity ranged from 89% for Crassostrea ariakensis to 72% for C. virginica. Curiously, HSP70 gene and cDNA sequences from M. galloprovincialis, deposited later (03 and 27 May), show only 73% identity with the present sequence.
