AT1-receptor blockade by telmisartan upregulates GTP-cyclohydrolase I and protects eNOS in diabetic rats.
2008
Several enzymatic sources of reactive oxygen species (ROS) were described as potential reasons of eNOS uncoupling in diabetes mellitus. In the present study, we investigated the effects of AT1-receptor blockade with chronic telmisartan (25 mg/kg/day, 6.5 weeks) therapy on expression of the BH4-synthesizing enzyme GTP-cyclohydrolase I (GCH-I), eNOS uncoupling, and endothelial dysfunction in streptozotocin (STZ, 60 mg/kg iv, 7 weeks)-induced diabetes mellitus (type I). Telmisartan therapy did not modify blood glucose and body weight. Aortas from diabetic animals had vascular dysfunction as revealed by isometric tension studies (acetylcholine and nitroglycerin potency). Vascular and cardiac RO…
– studium archeologiczno-historyczne
2020
Byczyna is today a small city, located in the northern part of the Opolskie Voivodeship. This stronghold is famous for well-preserved city walls, as well as for its history, especially that associated with the memorable battle of 1588. Of course, here is the clash between the troops led by Jan Zamoyski (1542-1605) and Archduke Maximilian III of Habsburg (1558-1618), which ended in the defeat of the latter. This event had a great impact on the history of the Polish-Lithuanian Commonwealth, as it pushed the Habsburgs away from royal power, as well as consolidated the rule of the Vasa dynasty. The Battle of Byczyna was an armed clash in which a representative of different nations met – Poles, …
Ikonoklazm – spór religijny czy polityczny? Leona III idea obrony czystości wiary i uniformizacji państwa
2017
Versican and collagen-III expression in bronchial and pulmonary muscular arteries in COPD patients
2015
Background: We demonstrated that COPD patients have thicker bronchial and pulmonary muscular arteries (MA) compared with never and non-obstructed smokers. The composition of the extracellular matrix of these altered MA has been not fully described. Objectives: To investigate the expression of versican and collagen-III in bronchial and pulmonary MA in the lungs of mild/moderate COPD patients. Methods: Eighteen mild/moderate COPD subjects (67±8y, FEV 1 : 67±15%pred), 14 non-obstructed smokers (64±8y, FEV 1 : 95±10%pred) and 11 never-smoking controls (CTRL) (51±12y, FEV 1 : 108±15%pred) were studied. Lung tissue obtained during surgery (ethical approval obtained) was used to analyze the expres…
Advances in haploidentical stem cell transplantation for hematologic malignancies
2016
One of the most important advances in allogeneic hematopoietic stem cell transplantation (allo-HSCT) is the use of alternative donors and cell sources, such as haploidentical transplants (haplo-HSCT) from family donors. Several approaches have been developed to overcome the challenging bidirectional alloreactivity. We discuss these approaches, including ex vivo T-cell-depleted grafts with megadose of CD34(+) cells, not requiring immunosuppression after allogeneic transplantation for graft-versus-host disease (GVHD) prophylaxis, and other strategies using unmanipulated T-cell-replete grafts with intensive immunosuppression or post-transplantation cyclophosphamide to minimize the GVHD. We als…
A blend of polyphenolic compounds explains the stimulatory effect of red wine on human endothelial NO synthase
2005
A high intake of polyphenolic compounds is likely to have beneficial effects on the cardiovascular system. Especially red wine is a rich source of polyphenols, and we have previously shown that French red wine upregulates eNOS, a protective enzyme in the cardiovascular system. The current study tested (poly)phenolic constituents of red wine for their ability to enhance eNOS expression (and the activity of a 3.5-kb human eNOS promoter) in human EA.hy 926 endothelial cells. Of the compounds tested, we found 3,4',5-trihydroxy-trans-stilbene (trans-resveratrol) to be the most efficacious stimulator of eNOS expression (and eNOS transcription), but this compound alone could not explain the total …
Prognostic and predictive factors in patients treated with ramucirumab (RAM) with advanced hepatocellular carcinoma (aHCC) and elevated alpha-fetopro…
2021
4146 Background: Elevated AFP in patients with aHCC is a poor prognostic factor with distinct molecular features, including high vascular endothelial growth factor (VEGF) signalling and increased angiogenesis. RAM, a human IgG1 monoclonal antibody, VEGF receptor 2 (VEGFR2) inhibitor, demonstrated improved survival vs placebo among patients with elevated AFP in the REACH-2 trial and is accepted as a standard of care for management of aHCC. We analyzed prognostic factors in patients with AFP ≥400 ng/mL and predictors of clinical benefit to RAM in an individual participant data (IPD) meta-analysis of the REACH and REACH-2 Phase III trials. Methods: Patients with aHCC, Child-Pugh A, ECOG perfo…
Stimulation of endothelial nitric oxide synthase by proinsulin C-peptide.
2003
There is increasing evidence for biological functions of human C-peptide. Recently, we have described that proinsulin C-peptide increases nutritive capillary blood flow and restores erythrocyte deformability in type 1 diabetic patients, whereas it has no such effect in non-diabetic subjects. The aim of the current study was to elucidate cellular mechanisms of this vasodilator effect in vitro by measuring the nitric oxide (NO)-mediated increase of cGMP production in a RFL-6 reporter cell assay and by demonstrating endothelial calcium influx with the Fluo-3 technique. C-peptide increased the release of NO from endothelial NO synthase (eNOS) in bovine aortic endothelial cells in a concentratio…
Dexamethasone lacks effect on blood pressure in mice with a disrupted endothelial NO synthase gene.
2003
Cushing's syndrome and systemic administration of glucocorticoids are associated with hypertension, but the underlying molecular mechanism is only partially understood. We have shown previously that dexamethasone downregulates the expression of the endothelial NO synthase (eNOS) gene in human endothelial cells and in the rat and that this may contribute to the blood pressure-raising effect of the steroid [Proc. Natl. Acad. Sci. USA 96 (1999) 13357]. In the current communication, we demonstrated that dexamethasone increased mean arterial blood pressure in wild-type C-57 Bl6 mice (eNOS+/+ mice), but had no effect on blood pressure in mice with a disrupted eNOS gene (eNOS-/- mice) derived from…
