Size measuring techniques as tool to monitor pea proteins intramolecular crosslinking by transglutaminase treatment

International audience; In this work, techniques for monitoring the intramolecular transglutaminase cross-links of pea proteins, based on protein size determination, were developed. Sodium dodecyl sulfate polyacrylamide gel electrophoresis profiles of transglutaminase-treated low concentration (0.01% w/w) pea albumin samples, compared to the untreated one (control), showed a higher electrophoretic migration of the major albumin fraction band (26 kDa), reflecting a decrease in protein size. Ligand-Binding Cooperativity Effects in Polymer-Protein Conjugation.

We present an electron paramagnetic resonance (EPR) spectroscopic characterization of structural and dynamic effects that stem from post-translational modifications of bovine serum albumin (BSA), an established model system for polymer-protein conjugation. Beyond the typical drug delivery and biocompatibility aspect of such systems, we illustrate the causes that alter internal dynamics and therefore functionality in terms of ligand-binding to the BSA protein core. Polystyrene nanoparticles were synthesized by emulsion polymerization of styrene. They were functionalized using the conventional surfactant N,N-dimethyloctylamine-N-oxide (ODAO), the tri-block copolymer (ethylene oxide)13(propylene oxide)30(ethylene oxide)13 (L64) and their mixtures. To this purpose, dynamic light scattering and calorimetric experiments were carried out and provided information consistent to each other. The L64 adsorption is Langmuir-type in the copolymer dilute regime and generates complex structures at larger concentrations. Dual Enzyme-Responsive Capsules of Hyaluronic Acid-block-Poly(Lactic Acid) for Sensing Bacterial Enzymes.

The synthesis of novel amphiphilic hyaluronic acid (HYA) and poly(lactic acid) (PLA) block copolymers is reported as the key element of a strategy to detect the presence of pathogenic bacterial enzymes. In addition to the formation of defined HYA-block-PLA assemblies, the encapsulation of fluorescent reporter dyes and the selective enzymatic degradation of the capsules by hyaluronidase and proteinase K are studied. The synthesis of the dual enzyme-responsive HYA-b-PLA is carried out by copper-catalyzed Huisgen 1,3-dipolar cycloaddition. Size characterisation of Au nanoparticles by ICP-MS coupling techniques

In modern nanotechnology the properties of nanoparticles (NPs) in different application fields is mainly dependent on their size and their size distribution. This paper aims to develop a reliable method for the size characterisation of Au NPs. Basing on the combination of two different separation techniques (liquid chromatography and gel electrophoresis), coupled on-line to inductively coupled plasma-mass spectrometry (ICP-MS), we study the separation of Au NP standards. Deciphering metal-induced oxidative damages on glycated albumin structure and function

Background: Metal ions such as copper or zinc are involved in the development of neurodegenerative pathologies and metabolic diseases such as diabetes mellitus. Albumin structure and functions are impaired following metal-and glucose-mediated oxidative alterations. Preparation and cytotoxic properties of goethite-based nanoparticles covered with decyldimethyl(dimethylaminoethoxy) silane methiodide

The present work describes the synthesis, physico-chemical and biological properties of the first water-soluble goethite nanoparticles covered with biologically active components: oleic acid and cytotoxic decyldimethyl(dimethylaminoethoxy)silane methiodide. The structure of initial goethite nanoparticles synthesized was proved by XRD analysis and the rough estimation of nanoparticles core size gave the value of 8 nm. The size of colloidal water-soluble nanoparticles, determined by dynamic light scattering, was within 19–35 nm.
