Vājās organiskās skābes spēj inhibēt mikroorganismu augšanu un metabolismu, tādējādi negatīvi ietekmējot biotehnoloģisko procesu efektivitāti. Veicot sūkalu fermentāciju ar raugu Kluyveromyces marxianus un novērojot acetāta uzkrāšanos fermentācijas vidē reizē ar augšanas un fermentācijas ātruma samazināšanos, tika nolemts izpētīt fermentācijās sasniedzamu acetāta koncentrāciju ietekmi uz šī rauga augšanu un metabolismu. In the present work, we analyse the sequences of the 5.8S rRNA gene and the two internal transcribed spacers 1 and 2 (5.8S-ITS region), obtained from 39 strains belonging to the species Kluyveromyces aestuarii, K. dobzhanskii, K. lactis and K. marxianus, K. nonfermentans and K. wickerhamii, to solve the phylogenetic relationships among these species and also to determine the possible genetic basis for the delimitation of the two currently accepted K. lactis varieties: lactis, including lactose-positive strains isolated from dairy products, and drosophilarum, comprising lactose-negative strains isolated from insects and plant exudates. Dairy Debaryomyces hansenii strains produce the antihypertensive casein-derived peptides LHLPLP and HLPLP

The ability of dairy Debaryomyces hansenii, Kluyveromyces lactis and Kluyveromyces marxianus strains to release the casein-derived antihypertensive sequences RYLGY, AYFYPEL, LHLPLP, HLPLP, VPP and/or IPP was examined. Yeast strains were grown in medium with casein as sole nitrogen source and the yeast casein hydrolysates (CSHs) were analysed by HPLC-MS/MS to search for the six antihypertensive sequences. Only LHLPLP and HLPLP were identified in CSHs and exclusively in D. hansenii Dh1 and Dh14 hydrolysates in which both antihypertensive sequences represented approximately 6 (CSH Dh1) and 10% (CSH Dh14) of total peptide content. Rauga Kluyveromyces marxianus acetāta tolerance

Maģistra darba mērķis bija novērtēt rauga Kluyveromyces marxianus iespējamos šūnu adaptācijas mehānismus pret acetāta izraisīto stresu. Fermentācijās ar laktozi saturošu substrātu un pievienotu 40 mM acetātu šūnu kultūrai tika novērota paildzināta lag fāze, kuras laikā palielinājās šūnu populācijas rezistence pret acetātu. Pētījumos tika novērota rauga Kluyveromyces marxianus DSM 5422 šūnu populācijas heterogenitāte, kas liecina, ka daļa no šūnām sākotnēji ir tolerantāka pret organisko skābju izraisīto stresu. Kultivējot raugu K. marxianus laktozi saturošā barotnē ar pievienotu acetātu, tika novērota iekššūnu vides pH palielināšanās. Iespējams, raugam K. marxianus vājo skābju izraisīta stre…
