Role of Th cytokines in thyroid autoimmunity

2007

Thyroid autoimmunity occurs when the immune system reacts against thyroid cells after targeting them like any other “not-self” antigen. To define a thyroid autoimmune disorder is often difficult even if the presence of auto-antibodies and/or autoreactive lymphocytes is a common feature. Although the pathogenetic mechanism is poorly understood, increasing evidences indicate that apoptosis is one of the key process that leads to thyroid auto-immunity. Afterward, Th cytokines have been considered the regulator factors of the cell survival in thyroid autoimmunity. Understanding the cell response from the perspective of type 1 or type 2 cytokines could clarify the pathogenetic mechanism controll…

MESENCHIMAL STEM CELLS IN INFLAMMATORY BOWEL DISEASES: CLINICAL EVIDENCES AND POTENTIAL INSIGHTS FOR THE CLINICIANS

2013

Mesenchimal stem cells (MSCs) have been used experimentally and clinically in the treatment of a wide variety of pathologies MSCs can be safely transplanted in autologous and allogenic ways as they are non-immunogenic, and consequently represent a therapeutic option for refractory connective tissue diseases, fibrosing diseases like scleroderma and fistulizing colitis like in Crohn's disease (CD). The immunomodulatory properties of MSCs have already shown promise when used as therapy for otherwise medically refractory CD. Accumulating evidence suggests that the properties may also be exploited of several other conditions. The currently available experimental and clinical data indicate that, …

NUTRITION IN IBD PATIENT'S: WHAT ARE THE PROSPECTS?

2015

Summary: Inflammatory Bowel Disease (IBD) is a chronic disorder characterized by a relapsing-remitting course, which alternates between active and quiescent states, ultimately impairing a patients quality of life. The two main types of IBD are Crohn's disease (CD) and Ulcerative Colitis (UC). In physiological conditions the gut is costantly exposed to various antigens, commensal microflora and pathogens and the inflammatory response is finely balanced. It is thought that a vast number of environmental risk factors may be implicated in the development of IBD, including smoking factors, dietary factors, psycological stress, use of non-steroidal anti-inflammatory drugs and oral contraceptives,…

Impact of Human Leukocyte Antigen Polymorphisms in Human Immunodeficiency Virus Progression in a Paediatric Cohort Infected with a Mono-phyletic Huma…

2014

Objective: HLA polymorphisms within the peptide binding pocket have been associated with rapid and slowprogression to AIDS, suggesting that the capability to present efficiently HIV-1 epitopes is crucial for the infection control. To minimize the effects of genetic background due to population coming from different geographic area and viral strain variability in the cohort, an analysis of all the polymorphisms associated with the HLA-A, -B and -DR alleles has been performed in a cohort of children with a monophyletic HIV-1 infection (CRF02_AG) during an outbreak in Libya. Methods: High-resolution HLA-typing has been performed in 58 children infected with a monophyletic strain of HIV-1: 26 L…

Slow-Ageing Diets

2019

Ageing is a complex, unavoidable phenomenon than cannot be exhaustively defined. However, it has been described as a process leading to death, characterized by a gradual decrease of the ability to adapt to stress, causing a decline in functional capacity. This event, marked by a progressive loss of physiological integrity of many interrelated systems, determines an increased risk of morbidity and mortality (Avery et al. 2014). With increase of the proportion of older adults worldwide (United Nations 2017), it is necessary to carry out prevention policy for a healthy ageing (HA). Good nutrition plays a significant role in determining the well-being of older people, and in delaying and reduci…

Tumorigenic and Metastatic Activity of Human Thyroid Cancer Stem Cells

2012

The recent discovery of CSCs as responsible for tumor development and progression has considerable therapeutic implications, since current cytotoxic drugs have a limited efficacy on CSCs and need to be replaced by target therapies. For this reason, much research effort has been addressed to identify and isolate CSCs as well as to improve understanding of the molecular mechanisms that control oncogenic process and chemotherapy resistance. Thyroid carcinomas are the most frequently diagnosed endocrine malignancy with a global increasing incidence. Data obtained during PhD course in Immunopharmacology showed that thyroid cancer tissues derived from follicular, papillary or anaplastic subtypes …

Phosphatidylserine liposomes reduce inflammatory response, mycobacterial viability and HIV replication in coinfected human macrophages

2021

AbstractChronic immune activation is the key pathogenetic event of Mycobacterium tuberculosis-human immunodeficiency virus (HIV) coinfection. We assessed the therapeutic value of phosphatidylserine-liposome (PS-L) in an in vitro model of M. tuberculosis-HIV coinfection. PS-L reduced nuclear factor-κB activation and the downstream production of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and IL-6 in bacille Calmette-Guérin-infected macrophages and of TNF-α and IL-1β in M. tuberculosis-infected and M. tuberculosis-HIV–coinfected macrophages. Importantly, a significant reduction of intracellular M. tuberculosis viability and HIV replication were also observed. These results suppor…

Distribution, function and predictive value of tumor-infiltrating γδ T lymphocytes

2013

Although γδ T cells are known to exert both tumor-preventing and tumor-promoting activity, there is still little information on the functional characteristics and clinical significance of γδ T cells isolated from the tumor site. In a recent study, we have investigated the distribution of different γδ T-cell populations in melanoma and their impact on disease outcome.
