Civic resielience : botanical gardens in North America, birth, development, and enviromental awarness

2023

The chapter explores from a historical perspective the birth of botanical gardens in North America, considered a significant part of the ecological network system and of the green and blue 'infrastructure'. This important system opposes geographical risk, which must be organically dealt with in programmes, plans and strategies that protect the landscape, prevent risks, and strengthen short-, medium- and long-term actions, also involving local Communities, integrating the vision to the different scales of intervention and projects. The Botanical Garden, seen here as an 'Open Work', has an important role not only because it preserves botanical specimens that come from all over the world, but …

A Thermophilic Tetramolecular G-Quadruplex/Hemin DNAzyme.

2017

International audience; The quadruplex-based DNAzyme system is one of the most useful artificial enzymes or catalysts; their unique properties make them reliable alternatives to proteins for performing catalytic transformation. The first prototype of a thermally stable DNAzyme system is presented. This thermophilic DNAzyme is capable of oxidizing substrates at high temperatures (up to 95 degrees C) and long reaction times (up to 18 h at 75 degrees C). The catalytic activity of the DNAzymes were investigated with the standard peroxidase-mimicking oxidation of 2,2'-azino-bis(3-ethylbenzothiozoline-6-sulfonic acid) (ABTS) by H2O2. The step-by-step design of this unique heat-activated G-quadrup…

Characterization of the Epithelial Permeation Enhancing Effect of Basic Butylated Methacrylate Copolymer—In Vitro Studies

2008

Membrane destabilizing properties and increased efflux of doxorubicin from liposomes caused by basic butylated methacrylate copolymer (BBMC), better known under its commercial trade name EUDRAGIT E, have been described in the scientific literature. Here, we investigated the effect of BBMC on suspended and filter-grown Caco2 cells with respect to apical-to-basal transport and membrane permeabilization using transport assays, trypan blue exclusion assay, measurements of transepithelial electrical resistance (TEER), confocal laser scanning microscopy, and transmission electron microscopy. The effect of inhibiting protein phosphatase 2A (PP2A) by okadaic acid was investigated by measuring TEER,…

Induction of apoptosis by arachidonic acid in human retinoblastoma Y79 cells: involvement of oxidative stress

2000

Arachidonic acid administration caused apoptosis in Y79 cells, as shown by typical morphological changes, phosphatidylserine externalization, chromatin condensation, processing and activation of caspase-3 and cleavage of the endogenous caspase substrate poly-(ADP-ribose)-polymerase. Arachidonic acid also caused lamin B cleavage, suggesting caspase-6 activation. Arachidonic acid treatment was accompanied by increased formation of the lipid peroxidation end products malondialdehyde and 4-hydroxy-2-nonenal, lowering in reduced glutathione content and in mitochondrial membrane potential. Inhibiting glutathione synthesis sensitized Y79 cells to apoptosis-inducing stimuli, whilst replenishing red…

Cytotoxicity and dentin composition alterations promoted by different chemomechanical caries removal agents : a preliminary in vitro study

2021

Background The use of chemomechanical agents for caries removal has been indicated as a non-invasive treatment option; however, their possible deleterious effects on the dental-pulp complex have been insufficiently studied. This study assessed the direct cytotoxicity of two chemomechanical caries removal agents (Brix 3000™ - BX and Papacarie Duo™ - PD) on pulp cells from deciduous teeth, as well as to assess the morphology and chemical compositions of the dentin surface after the application of these materials. Material and methods The cells were seeded (50,000 cells/cm²) in a culture medium (DMEM with 10% fetal bovine serum - FBS). After 24 hours, the BX and PD materials were added to 1:20…

Characterization of cobalt pigments found in traditional Valencian ceramics by means of laser ablation-inductively coupled plasma mass spectrometry a…

2008

Abstract In this work, a comparison of the performances of laser ablation-inductively coupled plasma mass spectrometry (LA-ICPMS) and portable X-ray fluorescence (XRF) spectrometry for the characterization of cobalt blue pigments used in the decoration of Valencian ceramics is presented. Qualitative data on the elemental composition of the blue pigments obtained using both techniques show a good agreement. Moreover, the results clearly illustrate that potters utilized different kinds of cobalt pigments in different historical periods. While both techniques seem suitable for the proposed task, they show different strengths and weaknesses. Portable X-ray fluorescence spectrometry is a cheaper…

The Role of Tissue Pressure and Bulk Flow in the Formation and Resolution of Cold-Induced Edema

1976

The mechanisms responsible for the dynamics of cold-induced edema, its spread and resolution, were examined. The findings indicate that bulk flow and not diffusion should be considered as the main mechanism for the edema spreading through the extracellular spaces of the white matter.

Determination of diphenhydramine hydrochloride by flow injection with Bromophenol Blue and turbidimetric measurement

1990

The study of a number of diphenhydramine-dye systems was carried out in order to determine the most suitable precipitate for the turbidimetric determination of diphenhydramine using flow injection (FI). The reagent selected was Bromophenol Blue. The chemical and FI variables were optimised. The calibration graph was linear over the concentration range 50-230 p.p.m. of diphenhydramine hydrochloride. A number of interfering substances were also investigated.

Staining mitochondria in Saccharomyces cerevisiae.

1969

After testing various procedures (amidoblack 10B, acid fuchsin-methyl blue, Luxol fast blue MBS-phloxine, toluidine blue O, Jams green B and pinacyanol), three stains can be recommended for staining both types of mitochondria (globose and threadlike) in the cells of Saccharomyces cerevisiae: (1) 0.1% solution of amidoblack 10B in citrate buffer (pH 3.0) for 10 min; (2) 0.01% solution of toluidine blue O in phosphate buffer (pH 6.0) for 30 min; (3) 0.01% solution of Janus green B in distilled water (pH 5.6) for 30 min. The latter stain is most specific because its staining reaction depends upon the action of the mitochondrial enzyme cytochrome c oxidase. Yet, low concentrations and short inc…
