IP 3 signalling regulates exogenous RNA i in C aenorhabditis elegans

2015

RNA interference (RNAi) is a widespread and widely exploited phenomenon. Here, we show that changing inositol 1,4,5-trisphosphate (IP3) signalling alters RNAi sensitivity in Caenorhabditis elegans. Reducing IP3 signalling enhances sensitivity to RNAi in a broad range of genes and tissues. Conversely up-regulating IP3 signalling decreases sensitivity. Tissue-specific rescue experiments suggest IP3 functions in the intestine. We also exploit IP3 signalling mutants to further enhance the sensitivity of RNAi hypersensitive strains. The article discusses the phenomenon of new media discourse permeating the language of written assignments produced in official school situations. The analysis of pupils’ essays concentrates on the colloquialisms transferred from social media discourse into the lexical layer of school writing. The author has assumed that the analysis will enable identifying educational barriers experienced by the new generation of pupils. The language of the texts under analysis combines the styles of written assignments, informal speech and of socialmedia discourse.

The purpose of this study is to describe the specificity of teaching Polish as a foreign language to Slovaks and Czechs (including the advantages and disadvantages resulting from language transfer), and to present the available teaching materials for Slovak and Czech students. The text includes an evaluation of the practical utility of different PFL textbooks designed for these groups and presents some suggestions for new teaching Solutions, such as the textbooks created at the Comenius University in Bratislava.

Mutant HRAS as novel target for MEK and mTOR inhibitors.

HRAS is a frequently mutated oncogene in cancer. However, mutant HRAS as drug target has not been investigated so far. Here, we show that mutant HRAS hyperactivates the RAS and the mTOR pathway in various cancer cell lines including lung, bladder and esophageal cancer. HRAS mutation sensitized toward growth inhibition by the MEK inhibitors AZD6244, MEK162 and PD0325901. Further, we found that MEK inhibitors induce apoptosis in mutant HRAS cell lines but not in cell lines lacking RAS mutations. In addition, knockdown of HRAS by siRNA blocked cell growth in mutant HRAS cell lines. Diacylglycerol kinases (DGKs) are a family of enzymes which phosphorylate diacylglycerol (DAG) to produce phosphatidic acid (PA), thus turning off and on DAG-mediated and PA-mediated signaling pathways, respectively.
