A semi-physiological two compartment pharmacokinetic model with two active metabolites (primary (PM) and secondary metabolites (SM)) with saturable and non-saturable pre-systemic efflux transporter, intestinal and hepatic metabolism has been developed. The aim of this work is to explore in several scenarios which analyte (parent drug or any of the metabolites) is the most sensitive to changes in drug product performance (i.e. differences in in vivo dissolution) and to make recommendations based on the simulations outcome. Hyperbolic subtraction method: Determination of the concentration of an analyte in the presence of an unknown interferent via spectral data.

A theoretical model to determine the concentration of an analyte in the presence of unknown interferents using spectral-type data is described. The method involves absorbance measurements at three wavelengths and the calculation of specific absorbances yielding a hyperbolic relationship between absorbance ratios and analyte concentrations. The concentration of the analyte of known spectrum can be determined in the presence of an interferent or mixture of interferents of unknown concentration(s) and spectra can be determined combining data for different sets of wavelengths. Electrochemical pH Control at Gold Nanowires

In this work, interdigitated arrays of nanowire electrodes are used with one array acting as the working electrode while the other is used to generate the required protons. Finite element simulations of the pH control electrodes were performed to provide insight on the generation and subsequent diffusion of protons. This informed the inter-tine spacing of the electrodes used.. This electrochemical pH control method was then used to enable the detection of analytes of interest.

X-ray analysis on the nanogram to microgram scale using porous complexes

X-ray single-crystal diffraction (SCD) analysis has the intrinsic limitation that the target molecules must be obtained as single crystals. Here we report a protocol for SCD analysis that does not require the crystallization of the sample. In our method, tiny crystals of porous complexes are soaked in a solution of the target, such that the complexes can absorb the target molecules. Crystallographic analysis clearly determines the absorbed guest structures along with the host frameworks. Because the SCD analysis is carried out on only one tiny crystal of the complex, the required sample mass is of the nanogram–microgram order. Cubic smoothing splines background correction in on-line liquid chromatography–Fourier transform infrared spectrometry

A background correction method for the on-line coupling of gradient liquid chromatography and Fourier transform infrared spectrometry (LC-FTIR) is proposed. The developed approach applies univariate background correction to each variable (i.e. each wave number) individually.
